True, even a two DOF system can exhibit chaotic... behavior, and a single cell is an at least an ~10^7 DOF system (i.e., protein molecules per cell). I still fail to properly convey to others the insane complexity I see in our microscopes, and why successes like AlphaFold only work because of the huge number of unnatural constraints placed on the training data. Case in point (below): lysosome dynamics over 12 min across a 200 x 70 um region in the head of a developing zebrafish embryo, color-coded by depth -- just one of 20k proteins at work. Our Cell Observatory Initiative is more important than ever, but while we have petabytes of the most mind-blowing data ever, we are still hamstrung by insufficient AI talent and compute -- two resources in great demand everywhere. Some microbes carry a protein, called SNIPE, that "chops... up" phage DNA as it's being injected into the cell. This is a new mechanism for phage defense! CRISPR–Cas and restriction enzymes also evolved to fight against phages, but they work by recognizing sequences. SNIPE works, instead, by sensing "touch." SNIPE is a protein with about 500 amino acids. After it's made by the ribosome, it latches onto ManYZ, two proteins which sit on the cell's inner membrane. (ManYZ is an importer; it brings mannose and other sugars into the cell.) Once attached to ManYZ, SNIPE sits and waits for an invading phage. Some phages, including lambda, actually infect cells by pushing their DNA through this ManYZ channel. Lambda uses its "tail" to reach inside the protein channel, basically, and inject its DNA. When this physical touch happens, though, SNIPE is waiting. As soon as the phage DNA starts entering the cell, and passes through ManYZ and SNIPE, it gets immediately destroyed. This means that SNIPE is the first phage defense system discovered, so far, that uses spatial positioning at the injection site to destroy invaders. But there are caveats, of course. If you untether SNIPE from ManYZ, such that it can freely diffuse through the cell, it will chew up the bacterium's genome. It is not a highly discerning nuclease! Also, SNIPE is not found in most bacteria. A prior pangenome study, which sequenced lots of different microbes, found that roughly a third of well-studied bacterial lineages had at least one member with a SNIPE-like protein. (For this paper, they just ported one of those homologs into an E. coli laboratory strain.) And finally, because SNIPE's mechanism is tightly tied to ManYZ, it cannot be used to defend against phages that enter the cell through different routes. T4 phages, for example, inject their DNA straight through the cell membrane and into the cytoplasm, without interacting with ManYZ. This is a nice basic science paper. Applications TBD. (Just remember that scientists figured out that bacteria had a phage defense system, called CRISPR-Cas, many years before it was repurposed into a gene-editing tool.) P.S. The video below shows how cells with the SNIPE gene (middle row) kill invading phages, and thus continue growing and dividing. Empty vector (top row) refers to bacteria carrying a plasmid with no SNIPE gene; this is a control group. And SNIPE E414A refers to cells which received a mutated SNIPE gene, where the glutamate at position 414 has been changed to an alanine, thus destroying the protein's nuclease activity. Artificial embryo samples aboard Tiangong Space Station for life... science research Space "artificial embryo" samples and other life science experiments delivered by the Tianzhou-10 cargo spacecraft have been installed into experiment modules aboard China's Tiangong space station, the Technology and Engineering Center for Space Utilization under the Chinese Academy of Sciences (CAS) said on Wednesday. The "artificial embryos" are stem cell-derived artificial human embryos. The embryos, though not true life forms and unable to develop into human babies, will help simulate the earliest stages of human development. Once the experiments are completed, the artificial embryo samples will be frozen in orbit and then returned to Earth on a future mission for side-by-side analysis with ground-based controls in laboratories on the ground. The bacterial flagellum looks like a simple tail, or... whip. But it’s actually a rotating motor, and perhaps the most sophisticated protein complex nature has ever evolved. In e. coli, these motors are capable of astonishing speeds; about 15,000 rpm. (The world record, according to one study, is for a Vibrio cell that was “clocked at 100,000 rpm by laser microscopy.) The flagellum propels the cell forward at speeds of 20-30 microns per second, or roughly 15 body lengths per second. If scaled up to the size of a cheetah, E. coli would *nearly* be the fastest land organism. The darting movements of a microbe were first observed in 1676 by Antony van Leeuwenhoek, a Dutch cloth merchant. Antony was delighted by the motion of his “animalcules,” writing: “I must say, for my part, that no more pleasant sight has ever yet come before my eye than these many thousands of living creatures, seen all alive in a little drop of water, moving among one another, each several creature having its own proper motion.” But Leeuwenhoek did not see flagella. He assumed, rather, that these animalcules must be “furnished with paws” instead. Christian Ehrenberg would not properly describe flagella until 1836. But amazingly, all the way up until the 1970s, nobody actually knew how the flagellum spun! In 1973, there were two competing models people argued over: the helical-wave (bending) model and the rotating (corkscrew) model. The first model suggested that the flagellum whipped back and forth, side-to-side, to propel the cell like paddle. The corkscrew model suggested that the whole flagellum instead spins around like a screw. In 1974, the corkscrew model finally won out. For two separate studies, scientists affixed flagella to glass slides using antibodies, and watched as the cells spun around and around like corkscrews. And finally, in just the last year, high-resolution structures of the flagellum have revealed a LOT more about its intricate assembly. The tail is made from ~20,000 self-assembling copies of a single protein, called flagellin. A “driveshaft,” or rod, spins the tail and is itself made of 26 protein subunits. Each “motor” in E. coli consists of 11 stators, each of which is made from 7 proteins.(Other types of cells have even more stators, and swim with much higher torques.) The flagellum spins when protons flow into the cell through tiny channels in these stators; akin to water running through a turbine. Each proton makes a small part of the stator change shape and push against the rotor, nudging it forward one step. With dozens of stators working at once, these nudges quickly spin the propeller. I'm writing an essay for Asimov Press about this now, and am really enjoying learning about the flagellum and its history. As the storms rocket into D.C. this afternoon &... tonight, please know that this was by design, with the foundation being laid for well over a week. First they peppered the entire U.S. with military contractors to ensure record temps & humidity. That provided the matter in the air that allows them to use NEXRADs and DEWs to create, steer, and intensity storms. Here's Lincoln, IL's NEXRAD this afternoon. It was juiced up by all the spraying of nanoparticles and use of DEWs. The blue dot tentacles of this (and every NEXRAD) extend well beyond the 100 nautical miles shown as the outer circle. Guildhouse was broken into and burglarized… …and it may... surprise many to know it was the 3rd burglary of 2024 and our 8th since we opened. These are not stats we are proud to be sharing, and in the past Guildhouse has not publicized these events for fear of inviting more theft. But this one was by far the most devastating, and it’s the reason Guildhouse has only been accepting credit cards for the previous 2 weeks. We’ve decided to share our experience this time because keeping quiet is clearly not working. On March 26 shortly after midnight, these clowns (pictured) used tools to cut through a sealed back door from the construction site behind Guildhouse. They then broke down the office door and stole our entire safe, along with various other property. For those unaware, small businesses like ours operate on very thin margins. Guildhouse in particular puts our mission of creating “a home away from home for the gaming community” above maximizing profit, striving to keep prices low and limiting the highly profitable events that otherwise interfere with gaming. Many of our fellow Downtown SJ businesses have also experienced the heartache and frustration of break-ins and burglary; the lowlifes who commit these crimes are too rarely held accountable. Despite the crime, we are determined to continue bringing our dream to life in #DTSJ and hope to work with SJ Downtown Association, City of San José and San José Police Media Relations to make meaningful safety upgrades to SoFA District. As some of us who live or have lived with them can attest, this often starts at 6am, or earlier…😊 A bright and happy “good morning” is a toddler greeting they don’t miss as they get excited about their day and want others to be excited too. And although many of us are not as awake as them during our mornings, it can help us, and those around us, to give that cheery “good morning” to everyone, including strangers. In a study looking at the behaviors of 60,000 adults, researchers from Turkey found that people who had these types of momentary interactions had increased wellbeing by establishing belonging. Lead author Dr. Esra Ascigil explained the reason for this as “Having a sense of belonging involves feeling like you are accepted and valued by other people – it is often considered a fundamental human need.” And we see this not just in the morning, but throughout the day as toddlers and preschoolers walk around malls and grocery stores greeting strangers much more than adults do. Lab-Grown Salmon Hits the Menu at an Oregon Restaurant... as the FDA Greenlights the Cell-Cultured Product | Margherita Bassi, Smithsonian Magazine The decision clears the way for the first cultured fish to join the small but growing alternative protein market While lab-grown chicken has been legal in certain U.S. states for a couple of years, the Food and Drug Administration (FDA) has just approved the first cultured fish for entry into the alternative protein club: salmon produced by the food tech company Wildtype. On May 28, the agency issued a letter stating it has “no questions” about Wildtype’s claim that its cell-cultivated salmon is “as safe as comparable foods produced by other methods.” This marked the final step in the FDA’s safety consultation process for the product, reports the Verge’s Dominic Preston. Emily Nytko-Lutz, an intellectual property lawyer who specializes in biotechnology patents, tells the Verge that seeking the agency’s pre-market safety consultation isn’t required for a company to sell its food, but it helps with marketing. In fact, Wildtype celebrated the green light by announcing a partnership with the James Beard award-winning chef Gregory Gourdet, who began serving the lab-grown salmon at Kann, his Haitian restaurant in Portland, Oregon, in late May. “Introducing Wildtype’s cultivated salmon to our menu hits the elevated and sustainable marks we want our menu to offer guests who share a similar value system to ours,” Gourdet says in the announcement. Lab-grown or “cultivated” meat has emerged as an alternative to killing animals for food. In terms of fish, the FDA’s decision comes as the seafood industry is suffering from pollution, climate change and overfishing, writes International Supermarket News. Meanwhile, seafood demand is projected to increase due to an ever-expanding human population and growing affluence. As such, lab-grown fish could lessen the burden on the fishing and fish farming industries, per Wildtype, as well as mitigate concerns about food contamination. To make their product, the food company’s scientists collect living cells from Pacific salmon and grow them in cell cultivators that mimic the inside of a wild fish—controlling factors like temperature, pH and nutrients, per their website. After harvesting them, the team incorporates plant-based ingredients to make the hunk of cells taste, feel and look like salmon fillets. They claim their final product has the same amount of healthy omega 3 and omega 6 fatty acids as traditional salmon, while avoiding the risk of mercury, antibiotics and parasite contamination. As noted by Popular Science’s Andrew Paul, Wildtype’s product is also notable because of its specific salmon cut, “saku”: the Japanese name for a block of seafood that can be eaten raw as sushi or sashimi. Currently, cultivated meat products are more expensive than animal products, and they’re sold at high-end restaurants. Wildtype’s salmon is only appearing on Kann’s menu on Thursday nights in June, but it will become available every day in July. I’ve prayed about whether I should post this publicly, and I have decided to do so because people need to know just how badly I am being targeted right now for simply being a lead journalist reporting on the Metcalf/Anthony case. On Sunday, August 24th, my home was swatted. Aside from the emotional distress this caused, especially as I was past my due date, we recovered and appreciated how well the department handled the incident. That same night, I went into labor. I labored at home from 11 PM until 3 AM, then headed to the hospital on the morning of August 25th. Then, at 12 noon on the 25th, while I was in the hospital in active labor, I received a phone call from my teenage son that Child Protective Services was at our door. I shouldn’t have to explain the extreme and immediate emotional distress this caused me and my family. Only cowards use the government to do their dirty work. This is the first time I have posted publicly about this incident. Kaidro has always been about the steady growth and long term play in the development of the intellectual property. Before launching the web 3 elements through out 2024 with 4 successful NFT launches, Kaidro had another lifetime in web 2 with the growth of the story and audience through the online manga on Webtoons exactly 4 years ago in 2020. We as the creators spent a good decade developing the world, storylines, art, and design as well as sharing the world alongside our audience of over 1+ million followers on TikTok. Since then, Kaidro has grown beyond a manga series to NFTs, an ARPG game, web game, audiobooks, merch, future animation and now the upcoming $KDR token.
