Some microbes carry a protein, called SNIPE, that "chops... up" phage DNA as it's being injected into the cell. This is a new mechanism for phage defense! CRISPR–Cas and restriction enzymes also evolved to fight against phages, but they work by recognizing sequences. SNIPE works, instead, by sensing "touch." SNIPE is a protein with about 500 amino acids. After it's made by the ribosome, it latches onto ManYZ, two proteins which sit on the cell's inner membrane. (ManYZ is an importer; it brings mannose and other sugars into the cell.) Once attached to ManYZ, SNIPE sits and waits for an invading phage. Some phages, including lambda, actually infect cells by pushing their DNA through this ManYZ channel. Lambda uses its "tail" to reach inside the protein channel, basically, and inject its DNA. When this physical touch happens, though, SNIPE is waiting. As soon as the phage DNA starts entering the cell, and passes through ManYZ and SNIPE, it gets immediately destroyed. This means that SNIPE is the first phage defense system discovered, so far, that uses spatial positioning at the injection site to destroy invaders. But there are caveats, of course. If you untether SNIPE from ManYZ, such that it can freely diffuse through the cell, it will chew up the bacterium's genome. It is not a highly discerning nuclease! Also, SNIPE is not found in most bacteria. A prior pangenome study, which sequenced lots of different microbes, found that roughly a third of well-studied bacterial lineages had at least one member with a SNIPE-like protein. (For this paper, they just ported one of those homologs into an E. coli laboratory strain.) And finally, because SNIPE's mechanism is tightly tied to ManYZ, it cannot be used to defend against phages that enter the cell through different routes. T4 phages, for example, inject their DNA straight through the cell membrane and into the cytoplasm, without interacting with ManYZ. This is a nice basic science paper. Applications TBD. (Just remember that scientists figured out that bacteria had a phage defense system, called CRISPR-Cas, many years before it was repurposed into a gene-editing tool.) P.S. The current state of the Islamic Republic is quite... strange. From a social restrictions perspective, it is utterly incomparable to when I was young or even 5 years ago. This goes far beyond people choosing to mass defy hijab laws - the restrictions against things like mixed dancing, concerts, music festivals, rave like atmospheres, drinking, recreational drugs, and all sorts of out of marriage sexual relationships are dissolved to a unrecognizable very low bar compared to the past. No it's not Turkey or Dubai (yet) and yes people still do get in trouble frequently (particularly businesses who host things - which is why you'll go to high end restaurants where everything is happening and they don't care but they'll immediately stop you if you take a picture out of fear it will go viral). But the cat is out of the bag. In every corner of the country the rules are flagrantly defied. Not just in cute underground parties at some well connected kid's house or some hippies doing drugs in a desert 3 hours from the city - no very much in the open, often advertised on social media, and not hidden at all (both videos are from the last month - the second video is in Shiraz). This was unthinkable just a few years ago. On the other hand the political atmosphere is currently closed at a level I don't remember other than the first few months after the 2009 election. It resembles the Islamic Republic of the late 80s more than what we've seen for most of my life. And from an international relations perspective all caution & restraint are gone and you see the most extremist elements (who ironically were kept in check by the older Khamenei who mistrusted them just as much as he mistrusted liberals) increasingly running the show, eagerly acting in a way that I struggle to describe in anyway other than delusional insanity. The economy is shattered to a level in which even the upper middle class are struggling to make ends meet, let alone the poor who are living through unthinkable conditions. This is a very different Islamic Republic from the "normal" baseline my generation is used to. Quality of life continues to nosedive at a horrendous level. Uncertainty is everywhere. External attacks are no longer a fear but a near daily reality. It is increasingly unclear who (if anyone) is in charge and what direction they are trying to steer the country in. The current situation might last for a bit as a transition state, but it is very difficult to imagine something this unstable, with so many contradictions, and this reckless, being durable. At some point very soon something will have to give. It can't last forever like this.show more

This grandad found a 500 year-old, 17ft deep well... under his living room in Plymouth, Devon, England. As well as, he also found a 16th Century BC, Bronze Age Sword and Coin from 1725 AD, inside the well :- The 70 year-old Colin Steer has spent last decade digging out well after he found it while redecorating living room of his home. He noticed a dip in floor while carrying out DIY, which led him to uncover well. While digging it out, Steer found what he thinks is the leather casing of a sword, which he believes could date the structure to 1500s. The 70-year-old said: "I found what seemed to be a leather casing of a sword when I was about 5ft into dig which leads me to believe that well could be medieval. "However it could date back to even earlier than that because we have no real way of accurately dating it at moment." He said that house was constructed in 1895 and he moved there in 1988 but it was 10 years ago while decorating that he saw the dip. "I immediately thought someone must have buried someone under there or that we had a sinkhole. "Since that moment, I decided I was going to dig it out and see what I would find. The hole is currently 17ft deep and there is about another 4-5ft that I want to dig out when I eventually get round to it. Since I began digging out well, I have also found a coin from 1725. In late 18th Century AD, during reign of George III, government issued large copper coins known as "Cartwheel" penny and twopence. An ancient sword was also discovered inside the well. Historically, sword developed in Bronze Age, evolving from dagger; earliest specimens date to about 1600 BC. The later Iron Age sword remained fairly short and without a crossguard. At the bottom of well the is about 4ft of water, so everything that I did out now will be wet and mucky stuff." Colin wants to go another few feet further down though before he finishes and although he is bringing up mud into his living room, he says that his wife Vanessa is understanding and is equally interested in the well. His plan is to then extend well wall up into his living room and use it as a coffee table which he thinks will make an attractive feature. He is also keen to find out why the well is actually there and consulting old maps have not given any answers. He said: "I would extend the walls of the well up through the floor and then add a round tabletop on top to make it into a coffee table. I think it would be a great feature for our front room. We still aren't really sure about why the well is there". The 17ft hole in the couple's living room is currently covered over by a sheet of Perspex and a trap door so that it is safe. Colin and Vanessa have grandchildren who love to look down the hole through the sheet of Perspex and they are very intrigued by unique feature in their grandparent's home. He added: "It is a good thing to show off to people as it is not something that you see every day. I have actually even tried some of water that is at the bottom of the well. It was really clear and tasted fine to me. I am going to get it tested for bacteria and if everything came back good then I could even bottle it up and sell it." According to the researchers, this well belonged to a wealthy or noble families, because only high status families could afford their own private well - most likely in the basement of your house, during medieval and late medieval times. Because, it was common for external wells to be poisoned in Medieval times, which is why castles would have them built internally. It could take decades to construct the well but it was worth wait to have your own clean water. To prevent well from collapsing, some were lined with stones, bricks or wood. Researchers also stated that, according to the preservation status of this well, it looks that the well was regularly maintained, to keep functional for long. Using four cores inside each fiber instead of one. Each core acts like an independent highway, and each band acts like an extra lane on that highway. Together, they’ve reportedly increased transmission capacity per core by nearly 50% and overall data throughput by up to 5×. This matters enormously for AI. Modern AI clusters move terabits of data per second between thousands of GPUs. The biggest bottleneck is often not the chips themselves, but moving data fast enough between them. If you can push 5× more data through the same physical cables, you can train bigger models faster and reduce network congestion. Why this is significant: • It shows multi-core + extended spectrum technology moving from labs into real-world commercial use • The system has already run over 35 km of existing telecom network • It could be especially useful for submarine cables and large-scale data center interconnects • China is also eyeing it for its “Eastern Data, Western Computing” project The deeper implication: We’re reaching the physical limits of how much data we can push through single-core fibers using traditional methods.
