(1) The human genome already contains more than enough diversity for natural selection - we generally don't need to worry about having a large enough population for new mutations. (2) Sometimes if selection is weak enough, it can be swamped by random genetic drift in small populations. But then it would be so weak that, even in a large population, it wouldn't make a difference on the timescales of human history. So population doesn't really matter either way.
