The Colosseum had a retractable roof, operated by a crew of sailors, almost 2000 years before any modern stadium. It was called the velarium: an enormous awning of canvas and rope that could be drawn across the open top of the arena to shade fifty thousand spectators from the Roman sun. It was so large and so complex that ordinary labourers could not manage it. The Romans brought in sailors from the imperial fleet, men who spent their lives handling rigging and sail, and stationed them at the top of the structure to extend and retract the canvas as the day moved. A building that has stood, roofless to our eyes, for centuries was in fact designed to be covered. That is the pattern with the Colosseum: almost everything about it was way more advanced than it looks today... Construction began around 72 AD under the emperor Vespasian. Once completed, it was the largest amphitheater in the Roman world: an elliptical structure of stone, concrete, and travertine, 189 meters long, rising as high as a modern fifteen story building. It could hold around 50,000 people and the staircases allowed that entire crowd to enter and leave with a speed that modern stadium designers still study. Beneath the arena floor lay the hypogeum, a hidden labyrinth of tunnels, cells, and machinery. Animals and gladiators waited there in the dark. Numerous trap doors opened in the wooden floor above them, and through hidden lifts and ramps a lion, a leopard, or an armed man could rise into the daylight as if from nowhere, in front of tens of thousands of people. The Romans knew that they had built something that would outlast them so completely that the Colosseum became, for the people who came after, a measure of the world's own endurance. In the 8th century, an epigram attributed to the Venerable Bede offered a prophecy that has never lost its allure: "As long as the Colosseum stands, so shall Rome; when the Colosseum falls, Rome shall fall; when Rome falls, so falls the world." Hello Hugh Thomas , let’s do it one by one, sample by sample, stone by stone. Whenever an artificial stone recipe of mine reaches a certain stage that’s the results are comparable to the natural version I’ll send a cast sample to you (with its mold) to analyze. The first one could be this perfectly chiseled fake limestone cube. As you can see you can’t fit a razor blade between the stone and the cheap silicone mold from Temu. This proves my micro chiseling skills. The recipe is public, you don’t even need my cube, you could create your own - but you won’t. So I’ll send this cute cube to you. The “secret” recipe is: I started from wood ash lye, also called potash, or K2CO3, by slowly dripping water through hard wood ash. Hard wood ash from your fireplace, yeah. Then I added slaked lime, and the chemical reaction produced fine, molecular level CaCO3 particles, fine limestone floating in the liquid. FYI: the secret chemical reaction is, and was in the ancient times as well: K2CO3 + Ca(OH)2 = CaCO3 + 2KOH Yummy! Next step: I let the limestone particles settle, and poured the KOH, potassium hydroxide solution above (that also formed in the liquid, see above) into the kitchen sink as it is a very good drain cleaner indeed. I added water and poured it into the sink several times, trying to clean the slurry from the KOH. So yes, the cube is from almost pure limestone, CaCO3 - with some residual KOH contamination. The KOH contamination can be reduced infinitely, I just stopped after 4 rounds. (Pro tip: put the slurry into a filter bag and put the bag into a mountain creek for an hour.) The next step is filtering and pressing the remaining liquid from the slurry. Because it’s a wet mess at this point. Pressure is a key factor to success! At least one thing is accurate from the universal canon: no pressure-no stone. How do I know? By trying and failing a few (dozen) times. So I poured the white limestone slurry into a sock 🧦 and pressed and squeezed it by hand - and that was JUST enough pressure for the magic to happen 😮 It looks like nature/god is on our side, everything is working as it was designed this way. Then I put the almost dry limestone paste into this Temu mold. Fun fact: it still contains a lot of water which will eventually be pressed out as the crystallization progresses, so we’ll need to get rid of that water through a hole, a nub. But there is no hole 🕳️ on this Temu mold so I poured the excess liquid from the top a few times. Hardening time=drying time=crystallization time : 1 week. The important lesson I learned from this experience is as follows: I tried to understand why this sludge turned into hard stone, because, as we were told, IT IS IMPOSSIBLE, and my understanding is this: wood ash lye does two different magics in parallel for this to happen: 1. It magically contains the right kind of contamination to facilitate calcite crystal formation and 2. The incredibly high pH (remember, 2 molecules of KOH are produced from 1 molecule of K2CO3!) in the liquid boost the calcite formation even more. ———- My only request from you and your colleagues concerning this analysis is full, 100000% transparency. And be honest. I know something you clearly don’t: you have lost this battle already. We need the data fast, not in 4 years or something. Transparency means sharing whatever you find here on X, without scientific drama and pullbacks. We don’t need a 947 pages scientific paper. We need the results, the data only. BTW: I already know what you will find: a “special type of natural limestone” with some minimal potassium contamination🤣🤣🤣 Now feel free to DM me, let this scientific analysis happen! A single E. coli cell, placed on a dish, will become 70 billion cells in just 12 hours. That’s exponential growth. But a new preprint shows that it's possible to engineer E. coli to grow linearly instead, where only one daughter cell continues dividing and the other stops. First, some context. In nature, there is a bacterium called Mycobacterium smegmatis (initially discovered in 1884 in ulcers scraped from syphilis patients.) M. smegmatis is weird because it divides asymmetrically. These cells grow only from one end, and all their cell wall biosynthesis machinery is located on that one end. So when the cell divides, one daughter gets this machinery and the other gets nothing. The daughter that gets the machinery can keep dividing immediately, but the other daughter has to remake all that machinery from scratch, so its growth is delayed. E. coli doesn’t grow like this. When it divides, it pinches in the middle and splits everything evenly. Enzymes, metabolites, and proteins get partitioned more or less randomly between the two daughters. For the new preprint, though, researchers engineered E. coli to behave more like M. smegmatis. Here is how they did it: First, they deleted a gene called cyaA, which encodes an enzyme (adenylate cyclase) that makes a molecule called cAMP. cAMP is SUPER IMPORTANT! It is a nutrient sensor that instructs E. coli to switch on genes that help it digest non-glucose carbon sources when glucose is scarce. Without cAMP, E. coli cells growing on alternative carbon sources will starve; they won’t know how to eat the food. Next, they added back a “split” version of the cyaA gene into the cells. In other words, they split the gene in two so that each half of the enzyme is made separately. Cells can only make cAMP, and thus eat non-glucose carbon sources, if these two halves come together. To facilitate that “coming together,” the researchers also fused the split cyaA proteins to sticky proteins that clump together, and to a fluorescent protein (to make it easy to track these molecules in the cell.) So now some interesting things start to happen if you grow E. coli on a growth medium lacking glucose. As the cell grows, its cyaA “halves” start clumping together into a giant ball. Inside the aggregate, the two enzyme halves come together and make cAMP. And when the cell gets big enough and divides, the clump of cyaA RANDOMLY goes to either daughter cell #1 or #2. The daughter that gets the aggregate (called PA+ in this paper) can keep dividing. The daughter that doesn’t (PA–) cannot. It still grows a few times — about four divisions — because it inherits some leftover cAMP from its mother. But after that, the metabolite is diluted away, and the cell stops growing. PA+ cells went through about 23 divisions on average before their aggregate decayed. And the population of cells, as a whole, grew linearly. This paper is cool because there are many applications where exponential growth is too unpredictable and, perhaps, unsafe. If you want to engineer bacteria to deliver drugs, clean up waste, or live in the gut, you don’t want them to double uncontrollably. This paper shows you can make them expand in a controlled, linear way. Alas, mutations could break this whole engineered system. A mutation that restores cyaA, for example, would give cells a new way to make cAMP. Mutations that make the aggregates split between daughters would break the asymmetry, too. But still, I really enjoy proof-of-concept engineering papers like this.
