Our #BlueSoup isolate is catalase positive! I compared a known positive cell line E. coli NEB Turbo vs our BS1-A isolate. Few drops of drug store H2O2 3% and watch for fizz. Catalase converts H2O2 into H2O + oxygen gas. Another clue as to the ID of this critter....

I didn't have a proper cat- cell line for a real neg control so this middle lane was to test if my ultra cheapo flint glass didnt have any embedded ions that would react with the H2O2 inorganically and give a false positive. @thuronyi is your S72 Vnat cat- ? Might use that if so!

As a non-scientist who stumbled in here from the internet, it’s interesting to see how many different tests you’ve had to do to figure this out. I guess I had just assumed it would be like birdwatching: field guide, microscope, done. Or have 🦠 pee on a stick & check for a “+”.

Phenotype can only get you so far, tbh. Sequencing is the true test but I'm an old soul and these sort of tests can be educational esp to folks who don't dabble in the squishy damp arts. We will do fancy sequencing and bioinformatics soon, these are just colorful edutainment :)

v exciting! i TA’d a microbiology course for med students this semester and they had a lot of fun doing tests to identify bacteria. this saga is so fun & exciting to me!! 😁😁

I don't know if you've already said, but do you plan to just sequence it sometime? I'd think just even some Sanger sequencing of RNAP should be enough to mostly ID, right?

Yeah @bielleogy is gonna do shotgun seq once she's recovered. I'm just enjoying the more accessible microbio in hopes of generating some content for educators to use if need be. I'll send it for nanopore via @seqcoast soon and Elinne and I will do hybrid assembly. Need blue first

Photorhabdus strain? Glows, catalase positive, blue pigment... what other tests have I missed. @Nick_Waterfield is your expert
