Our #BlueSoup isolate is catalase positive! I compared a known positive cell line E. coli NEB Turbo vs our BS1-A isolate. Few drops of drug store H2O2 3% and watch for fizz. Catalase converts H2O2 into H2O + oxygen gas. Another clue as to the ID of this critter....

I didn't have a proper cat- cell line for a real neg control so this middle lane was to test if my ultra cheapo flint glass didnt have any embedded ions that would react with the H2O2 inorganically and give a false positive. @thuronyi is your S72 Vnat cat- ? Might use that if so!

As a non-scientist who stumbled in here from the internet, it’s interesting to see how many different tests you’ve had to do to figure this out. I guess I had just assumed it would be like birdwatching: field guide, microscope, done. Or have 🦠 pee on a stick & check for a “+”.

Phenotype can only get you so far, tbh. Sequencing is the true test but I'm an old soul and these sort of tests can be educational esp to folks who don't dabble in the squishy damp arts. We will do fancy sequencing and bioinformatics soon, these are just colorful edutainment :)

I don't know if you've already said, but do you plan to just sequence it sometime? I'd think just even some Sanger sequencing of RNAP should be enough to mostly ID, right?

#WATCH | Hyderabad, Telangana: Six people, including TDP MP Putta Mahesh Kumar, tested positive for drugs in a raid by Police. EAGLE Force SP R. Giridhar says, "...We conducted a raid on a specific farmhouse where we found that one person was using a weapon. When we entered the farmhouse, our officers heard some sounds. They immediately rushed to the scene and captured one person who was holding a pistol. Another person also had some empty rounds. Officers took all of them, 11 people, into custody immediately. They conducted a drug test on them and 5 of them tested positive. We conducted all the procedural activities. They were taken to hospital for blood sample test. One person, in addition to the 5 people, tested positive. So, a total of 6 people were found positive. We handed them over to local Police, they will take further steps...While conducting test, we gathered information and came to know that one person is an ex-MLA and another is a sitting MP...Putta Mahesh (sitting TDP MP) tested positive in blood sample testing. We will produce them before the Court..."

Another YouTube Channel Idea: "E. coli Design." A whole series where you explain how to design DNA to change the behavior or appearance of E. coli cells. All you'd need is some equipment to transform cells, a microscope and camera, and some basic molecular biology equipment. Some ideas for modules: 1. The basics of a transcription unit; inserting GFP into cells and watching them glow. 2. Using a base editor to change GFP into another color? 3. Engineering E. coli to spin around in circles. 4. Engineering E. coli to swim faster, or to grow larger, or to partition their DNA unevenly, etc. ... and then end the series with the repressilator and blinking bacteria or something. India is known as the country of story-tellers and has the distinction of producing the highest number of films in the world. Our movies reflect our culture, traditions and are a major component of our soft power, whose influence has surmounted the barriers of time and boundaries. To further promote our film industry and bring a positive change in the lives of everyone associated with it, right from the spot boy to makeup artists to producers to distributors, the necessary amendment was brought into the bill. It will fight the menace of piracy, ensure minimum government interference as the need to renew the licence in every 10 years is done away with & categorisation & certification are also streamlined. In the #AmritKaal of our Independence, this law will enable our film industry to grow by leaps & bounds, bring employment to thousands of people & will play a prominent role in taking India to the top 3 economies of the world as envisioned by Hon'ble PM Shri Narendra Modi ji.

We’ve been holding public tests of a new Solo Matchmaking server, and experimenting with a ‘permadeath’ matchmaking option. We’ve also been doing more public tests with our ‘King of the Hill’ gamemode, allowing players to test their skill in team-based matches! The time between political decisions and actual damage to the enemy on the front lines, between the package's approval and our warriors' strengthening, must be as short as possible. This strength must be the one capable of truly changing the situation on the frontlines. Frontline air defense is just as important as protection for our cities and villages. Our long-range capabilities, artillery, and ability to expand our area of control are all important. Every day is important now, in terms of communication, politics, and logistics. And I am grateful to all of our friends in the United States, as well as our partners around the world, who see common opportunities and tasks in the same way that we do in Ukraine. Together, we must stop Russian terror by all means, limit Russia's terror potential, and force Putin to admit the obvious: this war will yield no results for him.
