IMO the most important scientific event this week was the publication of David R. Liu’s new Nature article 🧵👇in which he presented a new and promising Gene Editing modality named PERT - Prime Editing-mediated Readthrough of premature termination codons. This new modality - which uses Prime Editing to converted a regular tRNA gene into a highly optimized suppressor tRNA thus enabling permanent readthrough of premature stop codons, could potentially provide a one-time curative treatment for countless nonsense mutations which are responsible for almost one-third of all human genetic diseases! Here is an excellent lecture by Prof. David Liu - one of the most important scientists and innovators of our lifetime and the person who invented both Base editing and Prime editing - the 2 top leading modalities of Gene Editing, in which he throughly explains this huge scientific breakthrough. $PRME $BEAM

I would strongly advise every American leader and policymaker to listen carefully to David R. Liu - the most important scientist and innovator of our lifetime and the person who invented both Base editing & Prime editing - the 2 top leading modalities of Gene editing, when he describes in 60 Minutes the grave danger which the American BioTech ecosystem is facing due to insufficient federal funding. It seems the FDA has finally decided to cross the Rubicon and published a new and revolutionary regulatory framework which aims to expedite the regulatory approval process of new Gene Editing-based treatments - such as $BEAM Base Editing and $PRME Prime Editing. These new proposed guidelines are the pinnacle of a regulatory process which began with a Cell and Gene Therapy Roundtable - held last year by FDA former commissioner Marty Makary MD, MPH and the former director of the FDA’s Center for Biologics Evaluation and Research (CBER) Vinay Prasad MD MPH, in which researchers, industry leaders and scientists set together with FDA officials to bring new ideas to the table. During this fascinating meeting Prof. David R. Liu from Harvard University & the Broad Institute and most importantly the inventor of Base Editing and Prime Editing has made a closing statement which really caught my attention: “It’s not only about treating 1,000 rare disease patients but about building a modular system that turns rare disease treatment into a national engine for therapeutic innovation and which will secure America’s leadership in Genetic medicine for the foreseeable future.” It seems that finally Prof Liu’s call was answered!

It's a preliminary finding, but it may constitute a new gene editing mechanism that could have applications in gene therapy.
