A new mechanism for “RNA memory”! 😱 Thrilled to share another crazy paper from the lab (can’t believe we posted 2 in 2 days!), summarizing >10 years of research: Work on transgenerational inheritance of small RNAs in the powerful model organism C. elegans changed how we think about what’s possible in inheritance and evolution, because it allows the most heretical thing: inheritance of parental responses to the environment! However, it’s still unclear whether RNAs are inherited across generations in other animals, largely because the RNA-dependent RNA polymerases that amplify heritable small RNAs and prevent their dilution in C. elegans are not conserved in mammals. In this new work, an amazing collaboration with the Rink and Wurtzel labs, we show that planarians establish long-lasting and heritable small RNA–based gene regulatory states despite lacking canonical RNA-dependent RNA polymerases and nuclear RNAi machinery (that are required in C. elegans). You might say “they are both worms…” BUT planarians are evolutionarily very distant from C. elegans (flatworms vs. roundworms, diverged more than 500 million years ago), making this particularly surprising. These are totally different animals. We find that ingestion of double-stranded RNA induces sequence-specific silencing that persists for months and survives repeated cycles of whole-body regeneration. Even more strikingly, RNAi can be transferred between animals, echoing James V. McConnell’s controversial “RNA memory” experiments from the 1970s (his lab was targeted by the Unabomber terrorist Ted Kaczynski, who sent McConnell a bomb. This and other controversies ended this line of experiments…) Mechanistically, we find that the response transitions from a transient systemic dsRNA-triggered phase to a stable, cell-autonomous post-transcriptional “memory phase” maintained by antisense small RNAs. Using a new luminescence reporter (transgenesis is currently impossible in planarians), we show that silencing spreads along the targeted gene and identify a weird type of planarian small RNAs with untemplated polyA tails. RNAi inheritance without canonical RdRPs establishes planarians as a powerful system for studying RNA-based regulatory inheritance beyond C. elegans and raises the possibility that RNA-mediated inheritance may be more broadly conserved in animals, potentially even in mammals. Here’s a video of a planarian that is treated by RNAi against β-catenin and develops multiple heads instead of just one. This is one of the phenotypes that is inherited. Another phenotype is “loss of eyes” (which we show is not only inherited across multiple regeneration cycles, but can also be transmitted between animals in transplantation experiments). Every week, the average person ingests up to the equivalent of a 5-gram credit card in plastic. Microplastics are now thought to accumulate in the brain at 10 to 20 times the rate as other organs and, worse still, are 10-times higher in the brains of people who died from dementia. Most major organ systems are impacted. While the consequences of the pervasive presence of microplastics are still being understood, the microplastics themselves are carriers of harmful chemicals like BPA, phthalates, and PFAS. These harms are less ambiguous. Acting as a xenoestrogen, BPA, for example, has strong evidence that it affects multiple dimensions of child development ranging from the neurological to sexual and behavioral. Pregnant women with high BPA levels are six times more likely to have a child diagnosed with autism by age 11 compared to those with lower levels. Multiple lines of converging evidence demonstrate both reproducibility and even mechanism: BPA suppresses the aromatase enzyme, leading to anatomical, neurological, and behavioral changes in male mice that mirror aspects of autism spectrum disorder. In a study of 40 healthy young men, all were found to have microplastics present to some degree in their semen. Men with elevated BPA levels have lower sperm quality, reduced testosterone, and abnormal sperm morphology. The cardiovascular system is not spared. Microplastics have been found embedded in arterial plaques, and their presence correlates with a 4.5-fold increase in the risk of heart attacks and strokes. Cancer risk is another alarming aspect. Childhood exposure to phthalates is associated with a 20% increase in overall cancer risk and nearly a threefold increase in specific cancers like osteosarcoma and lymphoma. BPA's estrogen-mimicking properties have been linked to breast cancer development, promoting the growth of estrogen-sensitive cells and disrupting normal hormonal regulation. “In contravention to Judge Krupp’s preservation order, DA Morrissey’s Office gave the go ahead to Brian Albert and Brian Higgins to destroy or dispose of their phones.” ——Attorney David Yannetti, per the federal investigation Judge Krupp’s preservation order was issued back before the Fall of 2022, before #KarenRead’s case was transferred to Norfolk Superior Court. Preservation requests were subsequently re-examined and court orders reissued by judge Beverly Cannone upon Read’s June 2022 indictment and transfer to Superior Court. Presumably this means that the federal evidence shows that DA Michael Morrissey gave “witnesses” Brian Albert and Brian Higgins the go ahead to destroy evidence—their phones, back before Karen’s case moved over to superior court and judge Beverly Cannone ruled on the previously court-ordered preservation requests. Likely sometime in the Summer/Fall of 2022. This would also align with the fact that the Commonwealth refused to turn over the original Canton Police Department report to Karen Read’s defense counsel until October of 2022—over 8 months after she was arrested, at which point they’d surreptitiously altered the report in an effort to make Karen look more guilty. Notably, however, they also changed Brian Albert and Brian Higgins’ phone numbers on this altered report. This suggests that the timeframe when DA Michael Morrissey presumably gave both Brians the go ahead to destroy/dispose of their phones was sometime before October 2022, when this now altered version of the CPD report was finally produced to the defense. Why does this matter? On August 25, 2023, DA Morrissey put out a video statement personally vouching for the credibility of these same witnesses—Brian Albert and Brian Higgins. This is incredibly problematic for DA Morrissey. The mere fact that he apparently told “witnesses” they could destroy their phones is hard to overcome. How can DA Morrissey feign ignorance of the conspiracy to coverup Officer #JohnOKeefe’s murder after he himself gave “witnesses” permission to destroy their phones, or evidence? This fact alone makes him COMPLICIT. But beyond complicity, the fact that Morrissey then went on to make his exceedingly problematic video statement in August 2023 where he personally vouched for the credibility of these same “witnesses” thereby makes him a co-conspirator in the coverup of Officer O’Keefe’s murder AND in the framing of innocent woman Karen Read. What possible explanation could he offer for such actions and conduct? There’s no innocent explanation. “In contravention to Judge Krupp’s preservation order, DA Morrissey’s Office gave the go ahead to Brian Albert and Brian Higgins to destroy or dispose of their phones.” ——Attorney David Yannetti, per the federal investigation Judge Krupp’s preservation order was issued back before the Fall of 2022, before #KarenRead’s case was transferred to Norfolk Superior Court. Preservation requests were subsequently re-examined and court orders reissued by judge Beverly Cannone upon Read’s June 2022 indictment and transfer to Superior Court. Presumably this means that the federal evidence shows that DA Michael Morrissey gave “witnesses” Brian Albert and Brian Higgins the go ahead to destroy evidence—their phones, back before Karen’s case moved over to superior court and judge Beverly Cannone ruled on the previously court-ordered preservation requests. Likely sometime in the Summer/Fall of 2022. This would also align with the fact that the Commonwealth refused to turn over the original Canton Police Department report to Karen Read’s defense counsel until October of 2022—over 8 months after she was arrested, at which point they’d surreptitiously altered the report in an effort to make Karen look more guilty. Notably, however, they also changed Brian Albert and Brian Higgins’ phone numbers on this altered report. This suggests that the timeframe when DA Michael Morrissey presumably gave both Brians the go ahead to destroy/dispose of their phones was sometime before October 2022, when this now altered version of the CPD report was finally produced to the defense. Why does this matter? On August 25, 2023, DA Morrissey put out a video statement personally vouching for the credibility of these same witnesses—Brian Albert and Brian Higgins. This is incredibly problematic for DA Morrissey. The mere fact that he apparently told “witnesses” they could destroy their phones is hard to overcome. How can DA Morrissey feign ignorance of the conspiracy to coverup Officer #JohnOKeefe’s murder after he himself gave “witnesses” permission to destroy their phones, or evidence? This fact alone makes him COMPLICIT. But beyond complicity, the fact that Morrissey then went on to make his exceedingly problematic video statement in August 2023 where he personally vouched for the credibility of these same “witnesses” thereby makes him a co-conspirator in the coverup of Officer O’Keefe’s murder AND in the framing of innocent woman Karen Read. What possible explanation could he offer for such actions and conduct? There’s no innocent explanation. Excited to share our new work. Over the past decade, single-cell genomics has transformed our ability to map cellular systems. But a major question remains: Can we predict how perturbations reshape cellular trajectories over time? In 2018, we first showed that it is possible to predict cellular responses to perturbations — ranging from disease signals to chemical treatments — even in unseen contexts. In 2022, we introduced CPA (MSB 2022; NeurIPS 2022), extending this idea to predict responses to unseen chemical and genetic perturbations, including their combinations. Since then, the field of perturbation modeling has grown enormously. The community has pushed the space forward with many creative ideas and powerful models. It’s exciting to see how fast things are moving — even though many fundamental challenges remain. One of the biggest is that cells are not static. They move through trajectories during development, immune responses, and disease. Yet most current models still predict perturbation effects within a single state, rather than how early perturbations propagate across future states and reshape downstream outcomes. To address this, we developed PerturbGen, a trajectory-aware generative AI model that predicts how genetic perturbations reshape downstream cellular states. We explored this idea across three biological questions. First, in a human in vivo LPS immune challenge, PerturbGen predicted that perturbing a transient IL1B signal dampens downstream inflammatory programs in myeloid cells, with pathway changes reversing signatures observed in an independent IL-1β stimulation experiment. Second, in human hematopoiesis, PerturbGen predicted transcriptional responses to CRISPR transcription factor knockouts and enabled construction of perturbation atlases revealing lineage- and age-specific regulatory programs. These programs could also be linked to human genetics and blood diseases, including recapitulation of signatures associated with ETV6-related thrombocytopenia. Finally, we asked whether perturbation modeling could help improve complex tissue models. We built a dynamic perturbation atlas of human skin organoids to identify perturbations that could guideorganoid cells towardhuman fetal skin states. PerturbGen prioritized activation of Wnt signaling via GSK3β inhibition. Experimental validation confirmed the prediction: treatment with CHIR99021 induced stromal gene programs and shifted organoid fibroblasts toward transcriptional states observed in fetal skin stroma. Together, these results show how trajectory-aware perturbation modeling can connect gene perturbations to developmental programs, human genetics, disease mechanisms, and experimental interventions. More broadly, we think these point toward a future where single-cell atlases become predictive systems. As atlases expand across tissues, developmental windows, and modalities, models like PerturbGen could enable dynamic, virtual perturbation atlases— allowing us to simulate interventions, generate hypotheses, and design experiments before stepping into the lab. America's emergency oil is nearly gone, and it can never be fully refilled. The Strategic Petroleum Reserve fell to 298.7 million barrels this week, the lowest level since 1983. It dropped 6.1 million barrels in a SINGLE week. Against an authorized capacity of 714 million barrels, the reserve is now roughly 42% full. It held about 415 million barrels before the US and Israel attacked Iran on February 28. Iran moved to choke off the Strait of Hormuz, which carried nearly 20% of the world's oil, and a 172 million barrel release was authorized in March. Every outlet covering this is counting barrels. But the barrel count is the LEAST important number here... The reserve has no tanks anywhere in it. It is 60 caverns dissolved out of salt domes thousands of feet beneath Texas and Louisiana, at sites like Bryan Mound and Bayou Choctaw. To get the oil out, operators pump fresh water into the bottom of each cavern. The water is heavier, so it floats the crude up and out through the wells. But fresh water dissolves salt on its way to saturation. So every withdrawal eats the walls of the container. Siddharth Misra, a petroleum engineering professor at Texas A&M, said that the process leaves a "flatter, less stable roof" and "severely thins the critical salt pillars that separate adjacent caverns, greatly increasing the geological risk of a structural cave-in." The oil can be bought back at any price but nobody can put the dissolved salt back. The reserve was engineered for FIVE full drawdowns across its entire service life. It has literally executed dozens of releases in 40 years. Misra says that left "severe cavern deformation, accelerated the rate of massive salt falls from the ceilings, and significantly weakened the overall structural integrity of the aging reserve." Sandia National Laboratories has advised the reserve on salt since 1980. Its modelling now tells operators one cavern has three drawdowns left, another has five, and a third can NEVER be used again once emptied. One Sandia engineer summed the whole problem up by saying geology takes over and engineering stops mattering. Energy Department officials told the Government Accountability Office they are holding the reserve's infrastructure together with "Band-Aids," and that it is uncertain how long they will hold. Those same auditors found more than a quarter of the inventory was unavailable for drawdown as of December, because of construction and cavern outages. So the 298.7 million on paper is not what anyone can reach in a crisis. The number that matters more than the total: Misra says 70 million barrels is the hard floor that keeps the extraction pipes submerged in oil rather than water. The practical operating floor sits between 250 and 300 million barrels, and below 300 million the reserve loses its ability to pump at high speed. The entire point is moving crude to refineries fast. Sun-gazing to improve eyesight and eye-health? OK. Let’s break it down. See this video attached? It has more than 1 million views and close to 50K likes. In the video, a guy, who is the co-founder of The Satvic Movement, is trying to "naturalize" the Indians by claiming that everything scientific is “unnatural and synthetic.” He goes on to claim that he got rid of his “glasses” by looking at the Sun, a process known as sun-gazing – during dawn and at dusk which improved his eye health. He gives no scientific explanation for this, apart from a brightly lit well-edited video. Why do people require glasses? These are commonly due to disorders that affect the eye lens – to correct blurry vision that affects distance (nearsightedness – also called myopia), up close (farsightedness – also called hypermetropia), or both, along with difficulty in distinguishing certain shapes (astigmatism). Glasses work to correct your vision and are designed to alter the direction of light rays so that they shine correctly on your retina (the rearmost part of the eye where the light falls to create vision through multi-complex coordination of nerves), allowing you to see properly. These groups of disorders that affect the lens are called refractive errors and wearing spectacles are the easiest and most efficient and safest way to correcting vision in such affected people. In the video, a guy, who is the co-founder of The Satvic Movement, is trying to "naturalize" the Indians by claiming that everything scientific is “unnatural and synthetic.” He goes on to claim that he got rid of his “glasses” by looking at the Sun, a process known as sun-gazing – during dawn and at dusk which improved his eye health. He gives no scientific explanation for this, apart from a brightly lit well-edited video. Why do people require glasses? These are commonly due to disorders that affect the eye lens – to correct blurry vision that affects distance (nearsightedness – also called myopia), up close (farsightedness – also called hypermetropia), or both, along with difficulty in distinguishing certain shapes (astigmatism). Glasses work to correct your vision and are designed to alter the direction of light rays so that they shine correctly on your retina (the rearmost part of the eye where the light falls to create vision through multi-complex coordination of nerves), allowing you to see properly. On the second day of recruiting myths… Myth #2: “The name on the front of your jersey doesn’t matter.” There is some truth here. But people usually say this to make you feel better if you’re not on a team in the “recruiting bubble.” And yes of course there are always outliers. If you’re Montana Fouts, you could show up playing for the Butterflies and coaches would still be lined up drooling at the fence. But for most of us? The jersey does matter. Especially if you don’t have true 5-star size or can’t blow people away with elite metrics. I know this because I’ve been on both sides of it. Same player. Same work ethic. Completely different response from coaches. Why does the jersey matter more than people admit? First impressions. If a coach doesn’t recognize your club as one that consistently produces players at their level, you have to overwhelm them with performance and numbers just to get the same look. What matters even more than the jersey name is the coach attached to it. My last club, Rising, was brand new. But my coach, Steve Appel, had a lot of successful former players at top 25 programs. Coaches knew him. Trusted him. We were at the best fields and no one cared what was on the front of our jerseys. At my first tournament with that team, I saw more college coaches than I had in my entire recruiting journey before that. It’s true. College coaches aren’t recruiting a jersey. They’re recruiting you. But your jersey affects: •what fields you play on •who stops to watch •how risky you look on paper A well-known club with proven coaches comes with built-in credibility. Suspect in shocking church arson exposed in New Brunswick The man charged with burning down the largest wooden church in New Brunswick has, on his own Facebook page, a tribute to a radical friend who went to jail for fighting a pipeline. And on a video of that church burning — I’ve listened to it myself — a voice says that friend’s name, and then: “Your wish has come true.” I need to tell you how I know that, because it changes what I said last week about this church fire. I think it’s much worse now. On August 24th, at about 3:30 in the morning, the Saint-Simon Catholic Church — built in 1917 by the great-grandparents of the people who still live there — went up in flames. Four fire departments came. It was wood. It was gone in two hours. Only the chimney is standing. I flew to Moncton and drove three-and-a-half hours to see it for myself. I stood in the ashes with Claudette, the co-president of the parish council. The RCMP made a fast arrest — credit to them, that doesn’t always happen with church fires. Gilles Mallet, 68, from Saint-Simon itself. A parishioner himself! “He was at Mass with us,” Claudette told me. Charged with arson, remanded in custody. His bail hearing was put over to Tuesday, September 8th. Everyone in the village told me the same thing: one troubled man. Nothing bigger. I said on the show, fair enough — maybe. But after my report, an insider wrote to us through our website. He said he knew Gilles Mallet — from British Columbia, where Mallet had lived for years in an Indigenous community near Lillooet. Our source said he himself had left that community because of what was being said there about Christians and the Church. As in, Mallet had been stewing in anti-Christian propaganda. Our source asked us never to reveal who he is. We won’t. But Mallet wasn’t exactly hiding who he’d become. On Facebook he posted a picture of himself standing beside a war canoe. He’s wearing a red shirt that says “WARRIOR UP! — Ta’ah said.” Ta’ah is Amy George, Chief Dan George’s daughter, who coined “Warrior Up” in 2012 as the battle cry against the Kinder Morgan pipeline. Her son is Rueben George, who has led the Tsleil-Waututh fight against Trans Mountain ever since. Last year Mallet posted: “In honour of my friend Stacy Gallagher who’s in jail because of his fights against the pipeline.” He links to a video he made — “The Coast Salish Dragon, by Gilles Mallet (Baby Eagle).” Stacy Gallagher is a B.C. pipeline protester who was sentenced to 90 days in jail in 2021 for breaking the Trans Mountain injunction. And his cover photo, from this January: Mallet in a gold Inca headdress and cuffs. His caption: “Inka chief.” Then February, “Hawaiian Chief.” Then April, Peru. Obviously Mallet isn’t Peruvian or Hawaiian. But he “identifies” with Indigenous warriors fighting against Christianity. Now the video. It was posted to Mallet’s timeline a few days ago by someone in his circle. It’s the Saint-Simon church burning, shot from the road as the fire trucks pulled up. And the man holding the camera says: “Stacy Gallagher — your wish has come true.” The friend he honoured in March. The pipeline fighter. And as the church burned — 'your wish has come true.' Someone else in Mallet’s circle wrote this, over a video of him at a ceremony: “They manipulated him to do their evil work. He told them he was going to do such things.” Our source told us the same thing, too: that Mallet told people beforehand. “No one stopped him.” Being at a ceremony or wearing a slogan isn’t a crime. I don’t know what the “wish” was. But a man charged with burning a Catholic church to the ground wore a movement’s battle cry on his chest, honoured its jailed warrior, went deeper every month — and as the church burned, that warrior’s name was on somebody’s lips. That’s not a random fire. That’s a fire with a story behind it. We need to be back in New Brunswick on Tuesday — their court is open on Labour Day. Mallet is expected to apply for bail, and to enter a plea. I want to send Alexa Lavoie, because they speak French in Saint-Simon, and she will do much better than I did. But after my report, an insider wrote to us through our website. He said he knew Gilles Mallet — from British Columbia, where Mallet had lived for years in an Indigenous community near Lillooet. Her son is Rueben George, who has led the Tsleil-Waututh fight against Trans Mountain ever since. Last year Mallet posted: “In honour of my friend Stacy Gallagher who’s in jail because of his fights against the pipeline.” He links to a video he made — “The Coast Salish Dragon, by Gilles Mallet (Baby Eagle).” Stacy Gallagher is a B.C. pipeline protester who was sentenced to 90 days in jail in 2021 for breaking the Trans Mountain injunction. And his cover photo, from this January: Mallet in a gold Inca headdress and cuffs. His caption: “Inka chief.” Then February, “Hawaiian Chief.” Then April, Peru. Obviously Mallet isn’t Peruvian or Hawaiian. But he “identifies” with Indigenous warriors fighting against Christianity. Now the video. It was posted to Mallet’s timeline a few days ago by someone in his circle. It’s the Saint-Simon church burning, shot from the road as the fire trucks pulled up. And the man holding the camera says: “Stacy Gallagher — your wish has come true.” The friend he honoured in March. The pipeline fighter. And as the church burned — 'your wish has come true.' Someone else in Mallet’s circle wrote this, over a video of him at a ceremony: “They manipulated him to do their evil work. He told them he was going to do such things.” Our source told us the same thing, too: that Mallet told people beforehand. “No one stopped him.” Being at a ceremony or wearing a slogan isn’t a crime. I don’t know what the “wish” was. But a man charged with burning a Catholic church to the ground wore a movement’s battle cry on his chest, honoured its jailed warrior, went deeper every month — and as the church burned, that warrior’s name was on somebody’s lips. That’s not a random fire. That’s a fire with a story behind it. We need to be back in New Brunswick on Tuesday — their court is open on Labour Day. 2017 Capitulation Tour ⚔️ The plan to save the world has been in the making for many years. To the casual onlooker, the outworking of that plan may have looked a whole lot more sinister.
