Nice, economical protocol. To me the only really questionable step is the HotShot extraction - it doesn't sound like long-term DNA preservation is very good. Any new ideas on that?

This protocol is optimized for speed. If high quality DNA is needed for other purposes, one has to add the time for DNA extraction to the 6 hours.

Approximate total cost for setup minus typical lab materials?

Cost differs a lot between countries and is best determined for each country (but rough consumable costs are in the paper). The manuscript also disucsses how to trade off cost for time given that faster always means more expensive.
