Kearney Disinformation Correction Yesterday, Aidan Kearney attempted to attack the lack of canine DNA detected on John's clothing by tweeting two misleading claims. 1. Canine Swab Collection Kearney Disinformation: Michael Proctor was the source of DNA collected. Factual Information: The swabs were neither from Proctor's body nor collected by him. The swabs were collected by Maureen Hartnett at the MSP crime lab from John's shirt per instructions of Teri Kuhn at UC Davis and were received packaged appropriately. 2. Inhibitors Caused The Lack of Canine DNA Kearney Disinformation: There were inhibitors on John's clothing that masked the presence of canine DNA. Factual Information: UC Davis was asked to perform two tests: a nuclear DNA test which would be used to identify a specific animal and, if the nuclear DNA test failed to yield results, a mitochondrial DNA test called Meat ID to determine presence of DNA from a variety of species. Nuclear DNA is found in the nucleus of the cell and only 2 copies exist whereas mitochondrial DNA is found in the cytoplasm and is significantly more abundant with hundreds of thousands of copies. Nuclear DNA tests are far more accurate in determining the exact source, ie one dog from another dog, whereas mitochondrial DNA is less specific but will be able to determine if DNA from a specific animal species is present. In this case, the presence of DNA from 12 different animal species, including canine and pig, was tested. The results of the QPCR testing for nuclear DNA were that the swabs "did not indicate the presence of canine (nuclear) DNA, but did indicate the presence of inhibitors. This result is obtained when 1. Canine DNA is degraded, 2. The DNA is obscured by inhibitors, or 3. There is no nuclear DNA present." The less specific test using mitochondrial DNA, Meat ID, was then completed. This test showed no canine DNA and further, showed no inhibitors. In other words, had canine DNA been present, it would have been identified on the Meat ID mitochondrial testing since there were no inhibitors. As the Neanderthal genome is painstakingly sequenced, the archaeologists and biologists who study it will be faced with an opportunity that seemed like science fiction just 10 years ago. They will be able to look at the genetic blueprint of humankind's nearest relative and understand its biology as intimately as our own. In addition to giving scientists the ability to answer questions about Neanderthals' relationship to our own species--did we interbreed, are we separate species, who was smarter--the Neanderthal genome may be useful in researching medical treatments. Newly developed techniques could make cloning Neanderthal cells or body parts a reality within a few years. The ability to use the genes of extinct hominins is going to force the field of paleoanthropology into some unfamiliar ethical territory. There are still technical obstacles, but soon it could be possible to use that long-extinct genome to safely create a healthy, living Neanderthal clone. Should it be done? DNA has a shape, called a double helix, that makes it look like a twisted ladder. Each rung on the ladder is called a base-pair. The rungs are made up of a pair of chemicals called nucleotides--adenine, thymine, cytosine, and guanine, which are usually referred to by their first initials. The sequence of the nucleotides in the DNA determines what genes an organism has and how they function. Max Planck Institute initially chose 454 because it had come up with a way to read hundreds of thousands of DNA sequences at a time. Genome-sequencing technology is advancing at a rate comparable to computer processing power. Putting the fragments themselves in order can be a little tricky. "At first glance, it's just this completely random assemblage of As, Ts, Cs, and Gs," says Irzyk. "But it turns out there are patterns and motifs, and sometimes these are very specific to a group of organisms." For the Neanderthal sample, the human and chimpanzee genomes were used as references for checking the sequence. Working with ancient DNA can be much more problematic than sequencing genetic material from living species. Within hours of death, cells begin to break down in a process called apoptosis. The dying cells release enzymes that chop up DNA into tiny pieces. In a human cell, this means that the entire three-billion-base-pair genome is reduced to fragments a few hundred base-pairs long or shorter. The DNA also goes through chemical changes that alter the nucleotides as it ages--C changes into T, and G turns into A--which can cause the gene sequence to be interpreted incorrectly. In the case of the Neanderthal sample, somewhere between 90 and 99 percent of the DNA came from bacteria and other contaminants that had found their way into the bone as it sat in the ground and in storage. Issues of paternity have shaped our society for generations, especially in Zimbabwe. They did not start today. I remember going to a funeral with my father in the 1990s, and as we sat around the fire the men began sharing stories. One of them has stayed with me ever since. A Zimbabwean doctor had married a nurse, and they struggled to have children. As usual, when there is no child, the blame was placed on the woman. She went to see her own doctor, who told her plainly that she was not the problem, and advised her to bring her husband for testing. The husband refused, as many Zimbabwean men tend to do. Their marriage eventually collapsed, with the man insisting he could not be the problem because he had a child with another woman before marrying the nurse. That woman, he said, was also a doctor, and he was raising that child. After the divorce, the nurse remarried another doctor, and she immediately fell pregnant. Because it was a tight medical community, the whole situation became a talking point. The ex-husband had loudly declared that his former wife was barren, yet here she was having one child after another, three in total, with her new husband. Meanwhile, the ex-husband remarried, and again there were no children. One day the ex-husband and his former wife met at a gathering. During a casual conversation, as people who once knew each other, she told him directly that he was raising a child who was not his. She told him that he had no capacity to father a child and that if he finally went for tests, he would discover the truth. She reminded him how he had insulted her and her parents, and how he had refused to listen. The revelation shook him. He went for tests, and it was confirmed that he was the problem. He had spent seventeen years looking after a child who was not his. The moral of the story is simple, and I always stress this when mentoring young people. When in doubt, check. In fact, even without doubt, check. A DNA test at birth saves you from future heartbreak. Many men are raising children who are not theirs. Some women know exactly what they are doing. Others genuinely do not know because their relationships overlapped before they settled down. In those cases, even the woman cannot be sure who the father is. The client of the analysis is also a private individual. The biologist Dr. Jürgen Kirchner. He has been one of the vocal critics of mRNA vaccines for years. He operates a website called Gene Vaccines. There he also advertises his books written under a pseudonym. His last one is called "Sullied." This man noticed cans of the same brand piling up in his yard so he set up a ring camera and noticed the same vehicle passing by and throwing these cans over and over again but he needed a closer angle so he could get an accurate description of the vehicle so he set up a GoPro, when he finally got the information he needed he tracked the guy down and had an honest conversation with the man and found out he was doing it because of his Trump flag, he went on to explain to the gentleman the ways in which they were more alike than different and gave him an out, if he apologized to him and his family and promised not to do it again to which the man obliged. Too many times we see these situations culminate in violence and that’s exactly what those that propagate this hatred want, it is refreshing to see someone flip the script and handle it like a MAN. Hopefully these gentlemen have a newfound respect for each other and we’ll find a way to be neighborly to one another.

This week I spoke one on one with her father John Ramsey while guest hosting Banfield - we discussed the frustrating journey to justice and why the Colorado Police Department has not invited independent investigative genetic genealogy labs to help solve the case … yet. I asked John Ramsey about his recent conversations with a new investigator about why they haven’t taken up ’s offer to help. He said “Well, we did. And basically what he said, Okay, look, there's some things happening that I can't tell you about, and don't think DNA technology is there yet. I totally disagree with that. I met the founders of Othram Labs, which is one of the cutting edge labs out there that'd be happy to help. I sent a letter to the chief based on my meeting with what I learned with Othram they said A) they don't destroy any DNA when they do testing on it anymore, which was one of the concerns. B) they have the ability to separate DNA, mixed DNA samples, which is one of the issues with the sample they got in 1997 they had an unidentified male DNA sample in early 1997 it was mixed, apparently, with the blood DNA that can, now technology wise, be separated. That's huge. And the third thing, they have the ability to do familial genealogy research, which has been wildly successful in solving Old, old cold cases. That's what we're asking the police to do. Go to one of these cutting edge labs, let them do new testing, and they need a new DNA sample. We have a DNA sample which was obtained in January 1997 of an unidentified male DNA. It's not in the format needed to do this familial genealogy research to be a new command, and that's why we need to do more testing. NEW: Police airlift a crocodile with the body of a 59-year-old businessman inside. Local police in South Africa say they suspected the crocodile ate the man after observing it from drones. They then shot it and airlifted it off. When they landed back on the ground, the crocodile was sliced open, and human remains were found. The remains are believed to be those of 59-year-old Gabriel Batista, who had previously been swept away in raging floodwaters. His ring was found inside the animal along with six other pairs of shoes, which authorities fear may belong to other victims. On Monday, Batista reportedly tried to drive over a flooded bridge to reach his hotel and bar, but he and his truck were swept into the water. He was then swept down the river, where crocodiles were waiting for prey. It is unclear if Batista was already deceased when he was eaten, but authorities believe some of his other remains may have been eaten by other crocodiles.
