Ever wonder why we decide to build a new microscope and how we go about it? For the last lecture this semester of the graduate microscopy class Na, Gokul, and I teach at Cal, I spoke on our developing iSOAR microscope, a swept HILO design with adaptive optics and structured illumination. iSOAR is designed to produce the petabytes of high resolution 5D videos of subcellular dynamics in zebrafish we need to train the multimodal vision models at the heart of our Cell Observatory Initiative. We still have some work to do, but the scopes are coming along nicely. It's an exciting time at the Observatory, so if you're experienced and looking for a job in zebrafish transgenics, ultra-high throughput image processing, or the development of 5D spatiotemporal foundation models for interpreting the insane dynamic complexity of living matter, we'd love to hear from you. 4DGT: Learning a 4D Gaussian Transformer Using Real-World Monocular Videos Abstract: We propose 4DGT, a 4D Gaussian-based Transformer model for dynamic scene reconstruction, trained entirely on real-world monocular posed videos. Using 4D Gaussian as an inductive bias, 4DGT unifies static and dynamic components, enabling the modeling of complex, time-varying environments with varying object lifespans. We introduced a novel density control strategy in training, which allows our 4DGT to handle longer space-time input while maintaining efficient rendering at runtime. Our model processes 64 consecutive posed frames in a rolling-window fashion, predicting consistent 4D Gaussians in the scene. Unlike optimization-based methods, 4DGT performs purely feed-forward inference, reducing reconstruction time from hours to seconds and scaling effectively to long video sequences. Trained only on large-scale monocular posed video datasets, 4DGT can significantly outperform prior Gaussian-based networks in real-world videos and achieve on-par accuracy with optimization-based methods on cross-domain videos.

Metal manufacturing is essential for all areas of the economy. Because of their strength, stiffness, and long-term durability, metal components are used in applications from appliances to construction parts and car body panels. Traditional metal manufacturing techniques include forming, casting, molding, joining, and machining. Sheet metal forming involves various processes where force is applied to a piece of sheet metal to plastically deform the material into the desired shape, modifying its geometry rather than removing any material. Sheet metals can be bent or stretched into a variety of complex shapes, permitting the creation of complex structures with great strength and a minimum amount of material. Sheet metal forming is the most cost-effective forming procedure today for manufacturing parts in large quantities. It can be highly automated in factories or, at the other end of the spectrum, manually operated in metal workshops for small series parts. It is a versatile, consistent, and high-quality procedure to create accurate metal parts with limited material waste. From metal cans to protective housing for hardware, parts created by sheet metal forming are found everywhere in our daily lives. How does an embryo reliably "compute" its form - "cell by cell" - using only local interactions and mechanics, yet produce a precise global body plan? I’m excited to share our Nature Methods paper "MultiCell: geometric learning in multicellular development", presenting #AIxBiology research led by Haiqian Yang and the result of a great collaboration with Ming Guo, George Roy, Tomer Stern, Anh Nguyen and Dapeng Bi. A long-standing challenge in developmental biology is to predict how thousands of cells collectively self-organize as tissues fold, divide, and rearrange. In MultiCell, we represent a developing embryo as a dual graph that unifies two complementary views of tissue mechanics with single-cell resolution: cells as moving points (granular) and cells as a connected foam (junction network). This lets the model learn dynamics from both geometry and cell–cell connectivity. On whole-embryo 4D light-sheet movies of Drosophila gastrulation (~5,000 cells), our model predicts key cell behaviors and the timing of events, including junction loss, rearrangements, and divisions with high accuracy, at single-cell resolution. Beyond prediction, the same representation supports robust time alignment across embryos and offers interpretable activation maps that highlight the morphogenetic "drivers" of development. The broader goal is a foundation for cell-by-cell forecasting in more complex tissues, and eventually for detecting subtle dynamical signatures of disease. It has Quantum Properties?! Egyptian blue (calcium copper tetrasilicate, CaCuSi₂O₆) is the world's oldest known articial pigment. What makes this anomalous is not the pigment itself but its recently discovered quantum-optical properties. It emits near-infrared light with an exceptionally high luminescence quantum yield of 10.5%, making it a promising candidate for biomedical imaging, telecommunications, and laser applications. Even when reduced to nano-sheets a thousand times thinner than a human hair, this luminescent quality is fully retained. The pigment also has a chemistry similar to high-temperature superconductors. Washington State University researchers recreated the pigment in 2025 and found that "with just small differences in the process, you got very different results" This suggests that the ancient Egyptians had refined and controlled production processes whose knowledge was lost by the Renaissance.
