We’re thrilled to share that our MERFISH+ preprint is now live on bioRxiv!👉 In this work, the Bintu and Zhu labs (UCSD) developed MERFISH+, a next-generation spatial genomics platform that combines genome-wide RNA and epigenetic imaging over a large field of view. By introducing acrydite-modified probes covalently anchored to hydrogels, MERFISH+ achieves remarkable imaging stability and enables >1,800-gene, multi-modal, and multi-month experiments. With this platform, they, together with the Chi lab at UCSD, profiled a whole developing human heart at 12 post-conception week with merely two slides, resulting in a total of 53 slides, 3.1 million single cells and more than 30 cell types. Building upon our previous 3D reconstruction and modeling framework, Spateo ( we reconstruct the 3D human heart that nicely captures the anatomical structure of the heart, including the intricate vasculature network. Sophisticated analyses provide a holistic view of an entire organ and enable systematic characterization of 3D cellular neighborhoods and transcriptional gradients of substructures such as the descending arteries. Furthermore, using a generative integration framework for spatial multimodal data (Spateo-VI), we harmonized these MERFISH+ transcriptomic and chromatin data to reconstruct a 3D spatially-resolved multi-omics atlas of the developing human heart, shared at and MERFISH+ thus sets a new standard for large-format, multi-omic spatial profiling, enabling holistic, 3D characterization of organs at subcellular resolution. A new way of working. And a scary one at that. Memory Store is one of a group of new kinds of AI-first companies that can turn you into a Fast Company. I’m using several of them on my desktop and they are a dramatically new way to work. It builds a memory for: 1. Your AI agents. 2. Any employee using it. 3. The company itself. I sit down with founder Diwank Singh Tomer, Diwank Singh Tomer, who both freaks me out as well as shows how AI can radically help workers as well as managers. First, why does it freak me out? Well, his AI watches nearly everything a worker does and keeps a “memory” of it. It watches your email. Your calendar. Your Slack. And a whole lot of other things. This can really freak out workers if “forced” on them. And leads to a whole new set of security issues companies need to consider before adopting these things. Such data about a company could give a competitor a HUGE advantage, if leaked. They would know how a company “thinks.” It really is a surveillance system for employees and the company itself. OK, now why would anyone ever use such a thing? Because it gives employees super powers. It makes them more productive. Shows workers a lot of things about themselves, and helps them work and stay on task. It also gives the company super powers. Institutional memory stays with the AI now, even if an employee dies or leaves. As companies move to “AI First” approaches, they will increasingly see the value in companies like Memory Store. Decreases errors and unnecessary costs. Which tells me everyone soon will be using systems like this. This is what the new way of working looks like. Unfortunately, Tonfa sucks in Nioh 3 for me, because Tonfa without Fluxes are half the fun :/ Tonfa really gives its best with Fluxes. They were one of my favorite weapons in Nioh 1 & 2, super fun to play, and now they feel really frustrating 😢 This basically is my almost only main complain about Nioh 3, otherwise it's a 11/10 game for me. The main issue why I simply can't enjoy Tonfa on Nioh 3 is muscle memory. My fingers are so used to do Fluxes 2, they want to do Fluxes 2 — and not being able to do it makes me mad. Especially because Tonfa animations and moveset are still very similar to the previous games, so my body sees the animation and instinctively tries to Flux… but Ninja Style can’t do Fluxes 😭 With weapons like Kusarigama, which got a heavy rework, this problem doesn’t exist: animations are different, so muscle memory doesn’t clash. With Tonfa, instead, everything looks familiar, but a core mechanic is missing. Honestly, they should have either: -heavily reworked Tonfa animations for Ninja Style (for example somthing similar to Ninja Gaiden Tonfa Moveset) or -kept Tonfa in Samurai Style with stances and Fluxes like in Nioh 1 & 2 I don’t want Tonfa to disappear from Ninja Style — some people might enjoy them there — but I really wish Tonfa were available in Samurai Style as well, so everyone could enjoy the weapon in their own way. Please Team NINJA, give us back our beloved Tonfa in Samurai Style too 😭

I taught my Primary 2 students magnetic fields, and how they help us determine magnetic north and bearings using a compass. At the later half of the class, the students did a small experiment that used a magnet and compass to test what we learned. This is what Nigeria can take away from this: 1. LANGUAGE: Using Chinese to explain magnetic fields was so easy for them to understand, and took less than 4 mins. They never have to worry about understanding the grammar or remembering new vocabulary if another language was used. Language skill is not for everyone, just like technical and creative skills. While you or your son can learn a new language fast, others might be more technically inclined. Nigeria should use native languages to teach primary school children right from the first day in school. With many native languages, autonomous regions with different official languages will be ideal, with an option for international classes which are more English focused. 2. CURRICULUM: Unless we are really oblivious, there is no way a Nigerian primary 6 student can compete with my primary 2 students. These are kids that first came to our school so tiny and sometimes crying, many of them don’t even know how to write their names. A Nigerian taught them most of what they know today in less than 2 years, and because we have a curriculum that is modern, targets what the students really need to know, and its always modified to reflect their realities, they have acquired so much knowledge about how the world around them works like electricity, engines, space, aerodynamics, mechanics and environmental protection in a short time. What they are learning is directly linked to where their economy is headed, so by the time they are in university, they can easily understand advanced practical and theoretical knowledge and be useful to their economic development after graduation. Our curriculums in Nigeria are old and mostly useless to the growth of our economy, so let’s do the needful. Don’t wait for miracles to change them for you. We must demand this change. 3. GROUP WORK: Students are encouraged to work in groups, divide tasks and roles and they mentored to understand their strengths and challenges and resolve conflicts. They learn to work with others who are different from them, giving them a diverse perspective and mindset about working together. Nigeria cannot succeed without all of us uniting and learning to work with each other, irrespective of tribe, religion, region and family background. When we win as individuals, we will still be surrounded by helpless people, but when we win as a society, everyone develops. 4. TEACHERS: Here in China, we are always happy and encouraged to learn new things and ways to teach our students. We are free from worrying about food, shelter and pocket money. We are paid more than enough, so we have enough time and mental space to throughly research information and knowledge, and also design useful and interesting lessons for our students. Nigerian teachers should be paid a minimum of 800k to 1 million Naira per month. Primary school teachers lay down the crucial foundations for our economic growth, development and future. It is one of the most important professions in the country and the compensation and benefits MUST be more than senators and representatives. Liking, commenting and retweeting are nice because they help spread the message, however what’s more important is us uniting as Nigerians with our common interest to ensure that our needs are addressed.

Soccer Headers Damage Brains Even Without Concussions, Large Study Finds | Carly Cassella, ScienceAlert The world's most popular sport is reckoning with serious health concerns. The largest study of its kind has now found that repetitively heading a soccer ball can negatively impact the brain, even in amateur players who don't report concussions. Among 352 amateur adult soccer players, those who took more than a thousand headers a year showed microscopic changes to the outer wrinkles of their brains, right behind their eyes, regardless of their age or sex. These players also performed slightly but significantly worse on memory and learning tests. "What's important about our study is that it shows, really for the first time, that exposure to repeated head impacts causes specific changes in the brain that, in turn, impair cognitive function," explains neuroscientist Michael Lipton at Columbia University. Lipton's lab has been leading research on how soccer heading impacts the brain for over a decade now. Contact sports, like American football, Australian rules football, and rugby, are also contending with the downsides of repetitive head trauma, but in these cases, the discussion is often framed as a concussion crisis typically reserved for professionals. Lipton's research at Columbia University suggests that even mild bumps to the head can add up, and it's not just professional athletes or those who report concussions who are affected. Previous studies by Lipton have shown alterations to white matter among amateur soccer players compared to swimmers. Meanwhile, other studies have found white matter changes even without a history of concussion. But linking these brain changes to clear alterations in cognitive function has proven tricky. Lipton and his team have now developed a novel method that detects damage in the outermost wrinkles of the brain – a tricky spot to study using diffusion magnetic resonance imaging (dMRI). This layer is called the cortical gray matter–white matter interface (GWI), and until recently, not much was known about it. Using their new imaging technique, Lipton and his team scanned the brains of amateur soccer players, who had played for at least five years and within the last six months. Those who said they took more headers showed significant changes in the GWI at the front of their brains – a spot consistent with the trajectory of a soccer ball during a header. The authors suspect that this injury site reflects a contrecoup force – a sort of bruise to the brain that occurs on the opposite side of the skull. Injuries to the GWI may have gone undetected or underestimated in other imaging studies, which is possibly why there are conflicting results on the neurological impact of soccer headers. A graduate student in Lipton's lab, Joan Song, developed a way to characterize what they were seeing in the MRI scans. "In healthy individuals, there's a sharp transition between these tissues," Song explains. "Here, we studied if an attenuation of this transition may occur with minor impacts caused by heading." Sure enough, the boundary between white and grey matter was fuzzier in those who took more headers. "It's very strong evidence that these microstructural changes are likely to be a cause of cognitive deficits," Lipton says. Further investigation is needed, but the findings suggest that the GWI is a good place to image in future studies on soccer heading impacts. It may even be associated with disorders like chronic traumatic encephalopathy (CTE). Read more:

First impressions on Muse Glimmer! It's incredibly fast for a dense model, currently running an average of 208tps with a max of 274tps on a single 5090 with their DFLASH config. Comparatively, though, both using Open Code, Qwopus Coder (with thinking off) produced a much better shark survival game than the one I got from Glimmer. Meta's new dense model is currently just lacking some HTML canvas taste, but this is something that can be added via SFT as long as the model is stable and capable from a back-end programming perspective. And it seems to be, without a doubt. The big kicker here is that I ran this at extra high thinking, and it did not take long at all to run. Our current local leader, Qwen 27B 3.6, has a tendency to overthink, but with glimmer, that is not the case. Right now, my recommendation for general local programming (Apps, Games, Websites, Visual Tools) in this class is still Qwopus Coder with thinking disabled, or Qwopus Fusion with thinking enabled. Of course Shark Survival is a very basic domain-specific test, but I find that the result scales very well across many domains. If we're going to be shipping apps generated entirely locally, visual taste is somewhat of a bare minimum requirement, solely in my opinion, and Qwen's models in this class offer significantly more at the moment. That's actually why I initially started getting into finetuning with Qwen 3.5, they were the first base that was able to do really good front-end with some opus-trace fine-tuning. Qwen 3.6 has taste even in the base model, and we know Qwen 3.8 is going to blow us all away! Regardless, this looks like a very tempting new base model. I've started this private fundraiser to support our Kharkiv team, but it's not moving too well. We have only raised $1,000 out of our $12,000 goal. We have a small team in #Kharkiv that operates a warehouse, a repair workshop for engineering equipment, a machining shop for making parts, and a welding station for making anti-drone protective cages. My brother, vaycartana, has been running it for more than 20 years. My brother-in-law, Danya, was helping produce the anti-drone metal cages; however, he was mobilized to the National Guard 3 weeks ago, so we are now short one worker. Additionally, our main welder, my good friend Tolya, recently cut his arm tendon while using an angle grinder tool and is now recovering. They are working 12-hour shifts under tremendous stress and daily bombardments, repairing excavators, welding cages, and producing parts for our defenders. As you can see, the conditions are really harsh. The roof is leaking, the floors are rough, and daily bombings and a lack of personnel keep them struggling. The least I could do is run a fundraiser to fix and insulate their roof and install concrete, safe floors that can handle the excavators, to keep them warm and safe, especially during the winter. Their work saves millions of dollars of equipment and countless lives of Ukrainian defenders. The warehouse logistics they provide have always been free. Daniel reveals how he bought his $220,000 dream G Wagon with kids channel on YouTube Jack: “What's the craziest way you've made money on YouTube?” Daniel: “I just wrapped up running a channel around like 3 weeks ago for a new YouTube video I made. Essentially, the YouTube video was getting one subscriber equals $1 I can spend on my dream car. And so, I needed to get 220,000 subscribers to buy my dream car, which was a G Wagon for $220,000.” Daniel: “Basically, I ran this channel called Verusi. The videos were kids videos, and we would take these Russian creators' videos, they would make like really funny skits for like 5 year olds and we would put a voice over over them pretty much documenting what was going on in the video in the most 2 year old voice possible with the most simple wording possible.” Daniel: “I bought a pre monetized channel, which essentially meant that I bought the channel with already monetization enabled so that from the first video, we could generate revenue. Second video, 5 million views. Third video, 4 million views.” Daniel: “In the middle of the video, we would abruptly stop and we would be like, 'Subscribe if you want a pizza, like if you want a tomato, comment if you want an apple, and do all if you want your family to like live forever or some shit like that.' It was like really, really, really like brain rot, but it worked so well. We were getting like 100,000 subscribers a week.” Daniel: “I essentially realized that little kids don't have their own devices. They use their parents' devices to watch content. And so when a little kid is using their parents' device to watch a video, their parent is registered as a 30 or 40 year old. And so our entire audience demographic was 80% USA 40 year old viewers. And YouTube would essentially think that these are high trust advertisers.” Daniel: “We essentially tricked YouTube because getting kids to watch videos is super easy. It's not that hard. And our RPM was like 35 cents per video. And in the first week of that channel, we made $4,000. We paid an editor, I promise you, $5 per video. So, for a week, we uploaded seven videos. It costed us 35 bucks and we made $3,000 in our first week and got 150,000 subscribers.” Daniel: “I basically got 220,000 subscribers for the video, bought my car, and in the process made like $6,000 on that channel.”

For anyone dealing with cancer(s)... Here are a few helpful tips: 1st, don't have them "test" the tumor. Tumors are the body's way of protecting us from allowing the CA cells to traverse the body and mutate healthy cells. Once it's punctured for "testing", the cells travel and the beasties cause more issues. (Why not right? It's a $ making racquet.) 1A. CANCER IS A PARASITE. Start the Ivermectin (dosage 1am1 pm by bodyweight) & Fenbendazole (dosage 1 222mg pill/day for 6 days, skip the 7th to let the renal system rest). 2nd, STOP ALL SUGARS. CA loves sugar in ALL FORMS. 3rd, HYDROGEN PEROXIDE: H202 has the extra Oxygen molecule that CA can't stand. Put it on a cotton ball, or wash cloth for larger areas and rub all over skin. 3x/day, more-so when you're out of the bath & shower, as your pores are open and more apt to getting it into the bloodstream. The skin absorbs the HP, and kills the CA cells almost immediately. 4th, ALKALINE WATER: CA can't survive in an alkaline body. Start drinking alkaline water w/ lemon consistently thru the day. Lemon is an acid outside of the body, but the hydrochloric acid in your stomach converts it to an alkaline base. This travels through your blood stream and will help to eradicate the crud. 5th, DETOX BATHS: 1 cup Baking Soda, 1 Cup Borax, 1 Cup Epsom Salt, & Bentonite Clay. Draw the hottest bath you can muster... Soak as long as you can, as hot as you can, and it will help to detox the CA and the nano-particulates (gr@phine oxide), out of the body. Do this at least 2-3x a week. 6th, The BIG BOMBER: VITAMIN B17 which NO ONE wants ANYONE to know about! Apple seeds, Apricot, Peach, & Plum Pits, - ALL of it! You can get them on Cabalazon, or any other place online. The B17 vitamin has been removed from our knowledge, because: BIG pHARMa. 1/4teaspoon a day of the powder, or just eating a couple apricot pits thru the day, (like 3 or 4), will also eradicate the CA. I really hate the fact that non-suspecting people are dealing w/ all the bioweapon B.S. The turbo cancers are all a part of it. .. We have LEGIT CURES now. There's no longer a need for Chemo and Radiation which do nothing more than kill the good tissue. 9/10, the patient passes from chemo & radiation poisoning than the actual cancer. Before BIG pHARMa, CA patients were given 2 large bottles of peroxide, and a bag of cotton balls and told to use it daily and return in 2 weeks to find 100% of it was cured.
