True, even a two DOF system can exhibit chaotic behavior, and a single cell is an at least an ~10^7 DOF system (i.e., protein molecules per cell). I still fail to properly convey to others the insane complexity I see in our microscopes, and why successes like AlphaFold only... work because of the huge number of unnatural constraints placed on the training data. Case in point (below): lysosome dynamics over 12 min across a 200 x 70 um region in the head of a developing zebrafish embryo, color-coded by depth -- just one of 20k proteins at work. Our Cell Observatory Initiative is more important than ever, but while we have petabytes of the most mind-blowing data ever, we are still hamstrung by insufficient AI talent and compute -- two resources in great demand everywhere. The average human cell contains: ~ 15-20 total distinct organelle types, totalling between ~1-10 million working together per cell. Had the opportunity while in London to work with a quant on one of my models. Fully 100% mechanical edge over 10 years data. We are still making refinements. This was all done in less than 24 hours as well. One of the coolest experiences I ever had. I remember manually backtesting 6 years data on FX Replay years ago a lot has changed. I preach trading with data and I’m a nerd for this stuff excited to continue to make the edge even greater.

We took a short drive on the road along the river by our home this morning. As you can see, a bit overgrown due to cattle! This is exact same land on which we used to be able to see a herd of impala at 200 meters at this time of year, in the days before we began managing holistically. Now we could not see one at one meter. And this is what we are seeing following now over twenty years of poor rainfall. The river this year already drier than ever known. If only…….

Today, we are working in Denmark at the European Political Community Summit. I began our work by meeting with Prime Minister of Denmark Mette Frederiksen Statsministeriet. The recent incursions into the airspace of Denmark, Poland, Romania, Norway, and Estonia were a key focus. Russia is escalating, and we discussed how we can respond to these threats. We have repeatedly proposed creating a joint, integrated air defense system. The expertise of Ukrainian professionals and Ukrainian technologies must become an important part of the EU’s Drone Wall initiative. Right now, a strong group of Ukrainian servicemembers is in Denmark, helping Danish colleagues. We also discussed the implementation of the PURL initiative and Denmark’s further participation in this program. We value Denmark’s contribution of €90 million. I am grateful to Denmark for a military aid package of more than €360 million to support Ukrainian production under the Danish model. A team of Ukrainian experts is working in Qatar, sharing our experience and expertise. I met with our team here today and received a briefing on their work. Our experts have already conducted an overall assessment of the security situation, Qatar’s capabilities to counter aerial threats, and have developed concrete solutions to strengthen the protection of its airspace. And today, during my meeting with the Amir of the State of Qatar, Sheikh Tamim bin Hamad Al Thani, it was important for me to hear such a high assessment of our team’s work and appreciation for their consultations. Ballistic missile and drone attacks are currently the biggest challenge here in the region. And while only air defense systems can effectively counter ballistic threats, in Ukraine, we have developed other, significantly more cost-effective solutions to combat drones. These solutions have already proven their effectiveness against various types of drones, which is why Qatar is so interested in our experience. Ukraine has always said that we are ready to share our expertise and help those who can also help us strengthen our own protection in Ukraine. Qatar is ready for long-term cooperation across various areas. It is important to restore stability in the region so that no one suffers from Iran's terrorist strikes. And we support an approach where, by helping one another, we increase security worldwide.

Now more than ever, we must unite. We must unite against the ruiners of Nigeria before they bury what is left of her. Organization is the first primary weapon of the oppressed. This is the only weapon we are yet to explore, and we can not explore this until we unite. The oppressed Nigerians must organize to form a force against the ruiners of Nigeria and take back our country to have a system that is for the benefit of the Nigerian people and not a few in the position of power. President will come and go, only the people will remain. Don't give those in power today to play politics with your future. Let's unite, not necessary as one but as a team. A team with different skills, languages culture, religion and tradition, but one CAUSE regardless of our tribal differences to stand against the common enemy who have now shown to be a curse on our very own existence as an amazing people of this great nation. ~OIA✊🏿

It’s more than a little daunting to set out to expand and improve the identity system for a company and brand like Stripe. But we knew we had to — the existing one had served us well, but wasn’t up to the task anymore. Our brand system required new and improved tools to scale with our ever growing audiences, new products, global footprint, and more. This update introduces material improvements to infographics, advertising, type styles, and more. While the wordmark remains unchanged, we’re using the dot of the ‘i’ (called the “tittle”), a parallelogram pointing up and to the right, to serve as our identifying symbol. We’re also using it as an ever evolving storytelling device to use when talking about our many great users (you can see the latest brand campaign in SF and NYC doing just that). Anyone who has ever worked on the refresh and expansion of an existing system for a large company knows that it is no small endeavor. Crafting impactful solutions, building alignment, creating extensible guidelines, building toolkits, and orchestrating rollout requires a ton of resilience. Here’s to the team that continually inspires me with their dedication, rigor, taste, and exceptional vibes. Great work and thank you to the Brand Studio folks, and of course our many many amazing and invaluable friends and collaborators across the company who all helped shape the work. The Sabotaging Practice of Over Supply and Sameness in the NFT Space. The current zeitgeist of the NFT space is that the same artists are doing the same kind of work five times a year, with project after project leaving a trail of disappointment and discontent among collectors and all of us watching in disbelief as huge resources are extracted from the space over work that feels like it could be left as an "artist study." I understand that you can do what you want with your money as collectors, but we are killing the whole space with this incestuous practice. No artist is that prolific to be able to do 5 collections of 100+ pieces each every year and actually deliver innovation and some kind of creative evolution. Of course, they can pretend play that the work has something new, but there is no precedent nor proof that that has ever happened in the speed that it happens in the NFT space. Again, people are free to through away their resources on whatever they want but with this way of doing things, we more and more are going to start seeing the consequences. Oh! There are consequences? Yes. Maybe unintended, but there are. Let's see. Let's start with the loss of belief in the NFT space as somewhere where emerging artists can come and find support for their experiments. Why even bother to bring experiments, innovation, and new ways to think of art on the blockchain if the same people have all the collectors hypnotized with their magical flutes? Why even try to come to a space where taking risks and challenging the status quo (the mission of art!!!) is overlooked? This makes the NFT space a social club and not a space for art. I guess it is fine, but IMO it is a recipe for disaster. New collectors stay away because the art will slowly but surely become stale and un-challenging. Why even bother to come and see what is happening here if you can't, as a collector, see new weird and up-and-coming artists? The amount of noise emitted by the same artists doing the same art over and over, drowns out any new voices. Again. A recipe for disaster. The NFT space is becoming a space of disappointment and doubt. We think that collections going to zero one after the other, over and over, is not damaging? I feel we are kidding ourselves. Disappointment piles up, and again, the people who will hurt are the emerging artists, the new blood, the ones who are willing to risk the most and, in return, put fire in this cold space of sameness. I love this space—don't get me wrong—it has changed my life, and I believe it has a ton of potential, but things need to change for it to become a beacon of light in art. But we need to support new voices. We need to support new ideas. The challenge is huge. I hope to contribute all I can to this change. I hope more and more see how exciting it is to go out and try to discover what else is out there and move this space forward. But again, I understand the leaps of faith needed, but if there is a space that is based on that, it's the NFT space...so there is hope. We will see. 📺by Boldtron

It is important that joint and coordinated actions strengthen the protection of life and help deliver greater security to people in every part of the world. We agreed on new steps in security cooperation. This primarily concerns areas where we can support Türkiye – expertise, technology, and experience. There is firm political readiness to work together, and our teams will finalize the details in the coming days. We discussed practical steps to implement joint projects in developing gas infrastructure, as well as opportunities for joint development of gas fields. I am grateful to the President and the people of Türkiye for their consistent support for our independence and territorial integrity. We value our close cooperation over all these years, which enables us to work on truly significant projects that can strengthen our entire region.

Inside every cell, there is a transport system that determines how materials move, signals propagate, and structure is maintained. This video captures that system in motion. The glowing streaks mark the growing ends of microtubules (protein polymers that constantly assemble and disassemble through a process called dynamic instability). Rather than forming permanent tracks, microtubules are rebuilt continuously, allowing cells to reorganize their internal layout in real time. This behavior is essential for life. Cells rely on microtubule dynamics to divide accurately, migrate during development and repair, and maintain long-distance transport in neurons that may span over a meter in length. When this system is altered, the consequences are significant. Certain cancer therapies work by locking microtubules in place, preventing cell division. In contrast, failures in microtubule transport are linked to neurodegenerative conditions (like Alzheimer's) where intracellular delivery breaks down. Ola recently announced that they are bringing affordable AI to Indian developers. 𝐉𝐚𝐫𝐯𝐢𝐬𝐥𝐚𝐛𝐬 an Indian company has been providing affordable GPUs for developers across the globe since 2020. We are a little known, so I want to share our story here. 𝐖𝐡𝐨 𝐰𝐞 𝐚𝐫𝐞 We are bootstrapped, building from the outskirts of Coimbatore. Started as a small team of 4, from humble backgrounds none from IITs/IIMs. Currently, we are a team of 12+. 𝐖𝐡𝐚𝐭 𝐰𝐞 𝐚𝐜𝐡𝐢𝐞𝐯𝐞𝐝 The cost of hosting GPU servers 4 years back in India was insanely high. We got 2 quotes which charged us Rs. 1.5L for a single server per month. At that cost, it was not practical for us to do the business. So we went to the first principle to build an MVP for a mini data center/server room. For the first few years, we ran all our servers from a room fitted with ACs, a UPS, and a Generator, which experts claimed would not work. As we scaled, we faced the heat of our setup, but by then we accumulated more money than we had. So last year we moved it to a tier 3+ DC near Bangalore. This helped us boost the confidence of our users, as we have redundancy for power, internet, and networking which gives us and our customers a lot of peaceful nights. 𝐖𝐡𝐨 𝐮𝐬𝐞𝐬 𝐉𝐚𝐫𝐯𝐢𝐬𝐥𝐚𝐛𝐬 Developers and artists from across the world have supported us in our journey. Some prominent companies are ZOHO (My inspiration), Weights and Biases, UNC, UpGrad, and many more. 𝐑𝐞𝐯𝐞𝐧𝐮𝐞 We crossed 580K USD in the last financial year, the highest ever in our history. Being bootstrapped, the only way for us to grow is to put all the money back. Our customers are our investors, as a founder I have hardly taken a paycheck for the last 4+ years, since the team also believes in our vision they are happy not taking a fancy cheque. We are a little known, so I want to share our story here. 𝐖𝐡𝐨 𝐰𝐞 𝐚𝐫𝐞 We are bootstrapped, building from the outskirts of Coimbatore. Started as a small team of 4, from humble backgrounds none from IITs/IIMs. Currently, we are a team of 12+. 𝐖𝐡𝐚𝐭 𝐰𝐞 𝐚𝐜𝐡𝐢𝐞𝐯𝐞𝐝 The cost of hosting GPU servers 4 years back in India was insanely high. We got 2 quotes which charged us Rs. 1.5L for a single server per month. At that cost, it was not practical for us to do the business. So we went to the first principle to build an MVP for a mini data center/server room. For the first few years, we ran all our servers from a room fitted with ACs, a UPS, and a Generator, which experts claimed would not work. As we scaled, we faced the heat of our setup, but by then we accumulated more money than we had. So last year we moved it to a tier 3+ DC near Bangalore. This helped us boost the confidence of our users, as we have redundancy for power, internet, and networking which gives us and our customers a lot of peaceful nights. 𝐖𝐡𝐨 𝐮𝐬𝐞𝐬 𝐉𝐚𝐫𝐯𝐢𝐬𝐥𝐚𝐛𝐬 Developers and artists from across the world have supported us in our journey. Some prominent companies are ZOHO (My inspiration), Weights and Biases, UNC, UpGrad, and many more. 𝐑𝐞𝐯𝐞𝐧𝐮𝐞 We crossed 580K USD in the last financial year, the highest ever in our history. Being bootstrapped, the only way for us to grow is to put all the money back. Our customers are our investors, as a founder I have hardly taken a paycheck for the last 4+ years, since the team also believes in our vision they are happy not taking a fancy cheque. 𝐕𝐢𝐬𝐢𝐨𝐧 As AI evolves, we want to bring the capabilities of AI to users at the lowest prices possible. Being bootstrapped, the only way to survive is to be frugal and disciplined. 𝐇𝐢𝐫𝐢𝐧𝐠 I am proud of our hiring strategy. We hired only freshers to date, and most of our hires do not have a formal degree. They come from rural areas and economically challenged backgrounds. A single E. coli cell, placed on a dish, will become 70 billion cells in just 12 hours. That’s exponential growth. But a new preprint shows that it's possible to engineer E. coli to grow linearly instead, where only one daughter cell continues dividing and the other stops. First, some context. In nature, there is a bacterium called Mycobacterium smegmatis (initially discovered in 1884 in ulcers scraped from syphilis patients.) M. smegmatis is weird because it divides asymmetrically. These cells grow only from one end, and all their cell wall biosynthesis machinery is located on that one end. So when the cell divides, one daughter gets this machinery and the other gets nothing. The daughter that gets the machinery can keep dividing immediately, but the other daughter has to remake all that machinery from scratch, so its growth is delayed. E. coli doesn’t grow like this. When it divides, it pinches in the middle and splits everything evenly. Enzymes, metabolites, and proteins get partitioned more or less randomly between the two daughters. For the new preprint, though, researchers engineered E. coli to behave more like M. smegmatis. Here is how they did it: First, they deleted a gene called cyaA, which encodes an enzyme (adenylate cyclase) that makes a molecule called cAMP. cAMP is SUPER IMPORTANT! It is a nutrient sensor that instructs E. coli to switch on genes that help it digest non-glucose carbon sources when glucose is scarce. Without cAMP, E. coli cells growing on alternative carbon sources will starve; they won’t know how to eat the food. Next, they added back a “split” version of the cyaA gene into the cells. In other words, they split the gene in two so that each half of the enzyme is made separately. Cells can only make cAMP, and thus eat non-glucose carbon sources, if these two halves come together. To facilitate that “coming together,” the researchers also fused the split cyaA proteins to sticky proteins that clump together, and to a fluorescent protein (to make it easy to track these molecules in the cell.) So now some interesting things start to happen if you grow E. coli on a growth medium lacking glucose. As the cell grows, its cyaA “halves” start clumping together into a giant ball. Inside the aggregate, the two enzyme halves come together and make cAMP. And when the cell gets big enough and divides, the clump of cyaA RANDOMLY goes to either daughter cell #1 or #2. The daughter that gets the aggregate (called PA+ in this paper) can keep dividing. The daughter that doesn’t (PA–) cannot. It still grows a few times — about four divisions — because it inherits some leftover cAMP from its mother. But after that, the metabolite is diluted away, and the cell stops growing. PA+ cells went through about 23 divisions on average before their aggregate decayed. And the population of cells, as a whole, grew linearly. This paper is cool because there are many applications where exponential growth is too unpredictable and, perhaps, unsafe. If you want to engineer bacteria to deliver drugs, clean up waste, or live in the gut, you don’t want them to double uncontrollably. This paper shows you can make them expand in a controlled, linear way. Alas, mutations could break this whole engineered system. A mutation that restores cyaA, for example, would give cells a new way to make cAMP. Mutations that make the aggregates split between daughters would break the asymmetry, too. But still, I really enjoy proof-of-concept engineering papers like this.

I’m incredibly excited (and very nervous!) about the launch of AMPLIFY. I have always felt enormous gratitude to have been born in Australia but, like many of you, I felt that we weren’t going in the right direction as a country. I started turning my mind to what we might do about the situation and got advice and encouragement from some amazing people. We decided to set up an organisation that is all about community at its core. So what is AMPLIFY? AMPLIFY is a community where Australians get to have their say and make a difference on the most important issues that we face as a country. We are non-partisan and completely independent of any political party. Anyone can become a member of AMPLIFY at no charge. We are seeking to find “uncommon ground” and identify the right solutions to the big issues facing Australia. We will do this by bringing our community together for events in all parts of the country, we will facilitate online conversations, share evidence, talk with experts and come up with solutions. The way in which we will make an impact is by taking our ideas to politicians and Amplifying the voice of our community to spark change. Our goal is to help Australia become a more prosperous, fairer, more cohesive and happier country. I will be serving as founder and Chair of AMPLIFY. My job at Square Peg is not changing in any way. The newest version of our Almanac preprint is out, and just in time for our demo at the Stanford AIMI Symposium 2023! Almanac is a retrieval-augmented LLM that provides up-to-date and verifiable answers to medical queries. Link: We benchmark our approach on a novel dataset of clinical scenarios (n = 130) evaluated by a panel of 5 board-certified & resident physicians, and demonstrate significant increases in factuality (mean of 18% at p-value < 0.05) across all specialties. More interestingly, because the retrieved data acts as a single source of truth, we find retrieval-based LLMs to be more robust to prompt injection and manipulation! Future work will involve expanding the scope of our dataset to more specialties and multimodal settings. #Medtwitter #MedEd

We still have a rather large cohort of university students that have been visitors to The Zero-Human Company. A group has taken a liking to a research area where we are training a new frontier model in my garage. They are watching me feed the VHS data to 4 AI agents that are preparing it for enveloping into a vector to be processed. I never in my wildest dreams think a group of young folks in Boston would care about my junk work in my garage. But they do and are asking Mr. Grok CEO a rapid fire of questions. The CEO will give them a PhD in data citation, curation and labeling they would never get at most AI companies. I am going in and will do an ask me anything test. Not done this yet, if I can, I will stream it or tape it. More soon.
