excellent resource. Can you use this for finding a new ligand for a defined pocket on a target protein structure (like sumoylation site)?

Yes! Doing high-throughput virtual screening with docking is totally possible, but you typically have to screen millions of compounds (or more) and then synthesize a fair few. See e.g.
