A sneak peak of a complex and technically challenging... experiment that my lab developed: Super proud of PhD candidate Hannah Johnson for showcasing our Whole-gut spatial genomic analysis in #zebrafish. This video illustrates one landmark in the protocol after multiple rounds of sequential #HCR and 3D imaging in zebrafish larvae to reveal spatial expression of numerous mRNAs in the same specimen. Data from these imaging data sets are then computationally analyzed for spatial cell groups, spatially variable genes, and differentially expressed genes along 3D. We are leveraging this systems-level SGA to uncover unappreciated mechanistic insight at the cell and tissue levels into #ENS construction. Just had one of the worst drives I've ever... had in 5 years with FSD in the rain ... for some reason even though it limits you to Standard it would not stop getting into the left lane (and was capped at speed limit). And the left lane was flooded in spots... Tesla AI - the left lane obsession needs to end. At this point it's bizarre. Especially in these conditions. I manually had to move the car back to the right 10 times in 5 minutes. And overall ... I'm not going to lie I could not see anything. My heart rate was probably at 150 bpm. So I want to be an FSD but I don't want to be 3 ft from a barrier hydroplaning.. When the speed is limited in these conditions and it won't go the speed limit anyway - FSD NEEDS to stay out of the left lane!!!show more

THIS IS WHY I will always be a little... sick to my stomach when I think about the horribly managed ticket situation . Half the seats in my row (arguably the best in the venue) were empty and on top of that a few of those empty seats got filled by a trio who came 10 minutes late. Left halfway through the show and missed another 10 minutes, and the man with them sat for half of the show and shazammed/looked up the lyrics to almost ALL the songs including all her biggest hits😭😭😭.show more

Inside every cell, there is a transport system that... determines how materials move, signals propagate, and structure is maintained. This video captures that system in motion. The glowing streaks mark the growing ends of microtubules (protein polymers that constantly assemble and disassemble through a process called dynamic instability). Rather than forming permanent tracks, microtubules are rebuilt continuously, allowing cells to reorganize their internal layout in real time. This behavior is essential for life. Cells rely on microtubule dynamics to divide accurately, migrate during development and repair, and maintain long-distance transport in neurons that may span over a meter in length. When this system is altered, the consequences are significant. Certain cancer therapies work by locking microtubules in place, preventing cell division. In contrast, failures in microtubule transport are linked to neurodegenerative conditions (like Alzheimer's) where intracellular delivery breaks down. In the break between Pale Waves and Louis most of them were illuminated. All in the same light EXPECT for one. Which had a green light. •The VIP box right next to the green light was blacked out, by a curtain? Look at the box to the right of it. Every other dark box was very obviously just without light. • Louis kept looking in that direction, at least it looked like that. •Afterwards ,outside of the arena where there was the tour buses enter and the trucks unload. Pretty shortly fans are sent away from that side. Security said they couldn’t stand there anymore and got shooed away. Just when everyone left and no one seemed to pay attention, a completely blacked out VIP Van left the backstage area of the arena and drove by . This family was at the beach. They got there... early and set up in front so they could watch their kids play in the water. Then this family shows up and they set up in front of them. 🤯 There was no one to the left or the right of them. Clear as could be. 😳 A lot of people said they should have moved closer so no one could set up in front of them. But many others said if the beach was open and there was no one to the left or right of them then that is just plain rude. I always leave a little sand in front in case my kids want to play or we usually throw our boogie boards in front. I’d have been irritated and would have asked them to move. 💯 What do you think? Was it rude of the second family to take their view away and set up in front of them or is it the first family’s fault for not setting up closer so no one could be in front of them?show more

One of the things we've found is 3D printed... cloned plates to name one of many ways trucks get cloned across the industry. We see this in visual photos like Genlogs and we see this in the actual inspection, violation, and crash data. A truck can't be in CA and VA the same day. Why do we care? Israeli settlers entered the outskirts of Nablus this morning,... a week after a deadly gunfight took place in the nearby village of Tell, and again clashed with Palestinians, according to the IDF. The military says Palestinians hurled stones at the Israeli civilians, and one of the Israelis opened fire in the air. The IDF says it also received a report of several Israelis who "vandalized a cemetery" in the Nablus area. The settlers also passed through the outskirts of Tell during their hike. The hike, though areas A and B of the West Bank -- normally off limits to Israeli civilians -- was not approved by the army, but the military says troops "had been deployed in advance in the area ahead of the Israelis' arrival, were dispatched to disperse the gathering and worked to remove the civilians from the area." All of the Israelis eventually left the Nablus area, according to the army. The IDF says that later, Palestinians hurled stones at troops in the area, and the soldiers opened fire at a "main instigator." Palestinian media reported that one person was shot in the village of Juneid, near Nablus and Tell. "The IDF emphasizes that entry into Area A is prohibited and dangerous for Israelis, and constitutes a violation of the law," the army adds.show more

Facial reconstruction of a 4,300-year-old child from the Volga,... belonging to the genetically Yamnaya-like Catacomb culture. In the summer of 1989, an archaeological expedition of the Kuibyshev (Samara) State Pedagogical Institute investigated Kurgans of the burial ground near the village of Politotdelskoye (Kuzmina, 1990), a site comprising kurgans from different chronological periods. One of the Kurgans, dubbed Kurgan 4, contained an undercut grave (number 6). The oval entrance pit was oriented northeast–southwest and measured 1.78 m in length and 0.93 m in width. In the burial pit lay the skeleton of a child, around 7 years old. The soil beneath the skeleton and extending several centimeters around it was heavily stained with ochre. The coloration is especially strong in front of the torso, where the thickness of the stained layer reaches up to 10 cm. The bones themselves are lightly and generally evenly stained, with the hands being the most intensely colored. The skeleton was crouched on its left side, with the head oriented to the northeast. The skull is artificially deformed. The legs are slightly bent. The left arm is extended along the torso, while the right arm is bent at the elbow (the forearm lying across the torso, with the hand touching the left forearm). Along the left arm, in front of the face, by the left thigh, at the hands of both arms, on the left wing of the pelvis, and in the chest area, snake bones were found, heavily stained with ochre. Apparently, the snake bones belonged to several different snakes, although only one snake skull was discovered. The preservation of these bones is very poor, and their original arrangement is not fully intact. Animal bones were recovered behind the skull and near the left shoulder. Near the right mastoid process of the skull, a bronze pendant was found: a rounded spiral with one and a half turns, made from a rod of circular cross-section with flattened ends. Analysis of the most informative burials from the 1989 excavation at Politotdelskoye indicates attribution to the Volga–Don Catacomb culture.show more
